The Expressional Differences of Growth Differentiation Factor 9 and Bone Morphogenetic Protein 15 on sheep pre-antral follicles and their oocytes matured in vitro

Abstract: The study aimed to investigate the expressional changes of GDF9 and BMP15 on sheep follicle development and their in vitro matured oocytes. The micro-dissected technique was applied to isolate ovine pre-antral follicles. The expression of GDF9 and BMP15 on their pre-antral follicles and oocytes matured in vitro (0, 8, 16 and 24 h) were detected respectively by RT-PCR. The results showed that the cumulus cell expansion presented different alterations in morphology during vitro maturation of sheep COCs. GDF9 and BMP15 mRNA levels detected in pre-antral follicles were lower than those oocytes of different developmental stages as well as in vitro maturation. There were significantly transcriptional differences of GDF9 between preantral follicles and those different developing stages of oocytes, which suggested that GDF9 and BMP15 play an essential role during follicle development and in vitro maturation of sheep oocytes. All of these implied these two factors may independently or collectively affect the granulose cell proliferation and cumulus expansion.
