Comparison of Callus Induction, Somatic Embryogenesis and Plant Regeneration

from Different Clones of Hevea brasiliensis Anther

Abstract: In order to optimize the somatic embryogenesis and plant regeneration system of anther and provide a molecular breeding protocol for Hevea brasiliensis, callus induction, somatic embryogenesis and plant regeneration were compared under the same condition using anthers from six clones ‘Yunyan 73-477’, ‘Yunyan 73-46’, ‘Reyan 8-79’, ‘Yunyan 77-4’, ‘RRII105’ and ‘Reken 525’ of H. brasiliensis. The results indicated that the anthers from six clones were all able to dedifferentiate into callus tissues. But there were significant differences among different clones, ‘RRII105’ and ‘Reken 525’ showed the highest callus induction rates of 93.33% and 90.33%, respectively. The lowest differentiation rate was 31.33%, which was found in ‘Yunyan 77-4’. The somatic embryogenesis of different clones was significantly different, the highest differentiation rate was 76% which was found in ‘Yunyan 73-477’, and the clones of ‘Yunyan 73-46’ had no somatic embryo. ‘Reken 525’ and ‘Yunyan 73-477’ showed high plant regeneration rates of 75.38% and 54.35%, respectively, the clones of ‘Yunyan 73-46’ and ‘Yunyan 77-4’ had no regeneration plant.
