Establishment of Tissue Culture Regeneration System of leaf Lettuce

Abstract: In order to meet market demand, tissue culture technology is needed to lift the seasonal constrains of leaf lettuce production, and industrial mass production of the vegetable can satisfy people’ s daily need. In this study PS11 of leaf lettuce was used as test material, aseptic seedling was bred from the seeds; MS culture medium was used as basic culture medium with supplemental different concentrations of 6- BA and NAA. Through the callus induction, bud differentiation and rooting, a fast and efficient regeneration system of leaf lettuce was established. The results showed that: the best medium for callus differentiation was MS+0.2 mg/L 6-BA+0.4 mg/L NAA, the callus induction rate reached 100%, and the budding rate reached 71.6%.
