Distribution and Identification of a New Cytoplasmic Male Sterile (CMS) in Cultivated Rice

Abstract: The study aims to analyze the distribution of cytoplasmic male sterile (CMS) gene orf216 in cultivated rice, and to identify new resource of CMS. 824 cultivated varieties were analyzed with PCR amplification using special primer Sp1F/Sp1R of HL-sp1. The results showed that there were 9 accessions with HL- sp1. Then, the above 9 accessions were further analyzed by PCR amplification using CMS related gene orfH79, orf216 and Southern Blot. Results showed that three accessions (IR72892, IR43, IR73546) contained only orf216 no orfH79 in selected cultivated varieties. By marker- assisted selection (MAS) method, new cytoplasmic types related CMS have been achieved from 824 cultivated varieties. These results contribute to the study of gene function and the development of CMS line in rice in the future.
