Variance Analysis of Genetic Diversity of Potato with SRAP and SSR Markers

Abstract: This study was to understand the genetic diversity and genetic background of the main potato varieties in Ningxia, clarify the relationship among varieties, and provide the theoretical basis for potato breeding. The genetic diversity of 47 potato germplasm materials was studied with SSR and SRAP markers, and the results of these two maker systems were compared. The results showed that 180 polymorphic fragments were detected using 12 SRAP primer pairs, with an average number of polymorphic fragments per primer pair of 15. The PIC for SRAPs was from 0.2 to 0.43 with an average of 0.34. The genetic similarity coefficient of the 47 germplasm materials varied from 0.57 to 0.83 by SRAP markers. Whereas, 80 polymorphic fragments were detected using 15 SSR primer pairs, with an average number of polymorphic fragments per primer pair of 5.3. The PIC for SSRs was from 0.37 to 0.72 with an average of 0.51. The genetic similarity coefficient of the 47 germplasm materials varied from 0.60 to 0.97 by SSR markers. According to the pedigree information, the SRAP markers were more effective for genetic analysis of closely related lines. Potato varieties in Ningxia had low genetic similarity and distant genetic relationship.
