Optimization of Poly (Vinyl Alcohol)-Degrading Enzyme Production Condition in Bacillus amyloliquefaciens HK1

Abstract: The strain HK1 was isolated from environment, which could degrade polyvinyl alcohol. Using the I2-KI and boric acid staining method, it was proved that HK1 had intracellular and extracellular degrading enzymes, mainly extracellular enzymes. Optimization of culture conditions was carried out through mono-factor and orthogonal experiments. The results showed that: the optimum loading volume, inoculum density and culture temperature was 50 mL/250 mL, 6% and 30℃, respectively. The optimum external carbon source and nitrogen source was PVA and NH4NO3, respectively. The optimal fermentation condition was: PVA 2.5 g/L and NH4NO3 0.6 g/L at pH 7.0. Under the optimal fermentation condition, the production of degrading enzyme of strain HK1 was 225% higher than when it was cultured in former fermentation condition.
