Expression of GlyCAM-1 in Uterus of Artificially Induced Oestrous Sheep

Abstract: To investigate the changes in the expression of GlyCAM-1 in uterus of oestrous sheep which was artificially induced, expression changes of GlyCAM-1 on the 10th, 12th, 14th, 16th and 18th day after sheep oestrus were detected respectively by Real-Time PCR and Western blot. The results of Real-Time PCR showed that mRNA of sheep GlyCAM-1 increased on the 10th day, decreased on the 14th and 16th day, but increased again on the 18th day, which were in accordance with the results of protein expression detected by Western blot. Protein of 34 kD was detected by this method with specific antiserum. The transcriptional and expressional level of GlyCAM-1 on the 10th day was significantly higher than that of the other days (P<0.05). In addition, GlyCAM-1 mRNA expression on the 12th day reduced significantly (P<0.05), and then remained relatively constant, but significantly increased on the 18th day. There were no significant difference of protein expression among the 12th, 14th and 16th day (P>0.05), but there were significant differences among the other days (P<0.05). Protein expression of the 18th day increased significantly (P<0.05). Thus it could be concluded that during the 10th-18th day after sheep oestrus, the transcriptional and expressional level of GlyCAM-1 presented a decrease-increase trend.
