Isolation and Identification of Mycoplasma bovis FJ-HJ Strain

Abstract: One strain of Mycoplasma bovis was isolated from lung tissue of an infected beef cattle characterized by gasp and runny noses in Fujian Province. To identify this strain, colonial morphological observation, biochemical test, specific polymerase chain reaction (PCR), cloning and sequencing of 16S rRNA gene were conducted. The results showed that colonies of this strain, which was named as FJ-HJ, were like fried eggs with brown protrusion in the center. FJ- HJ could not ferment glucose, not hydrolyze arginine, and not decompose urea. Hemadsorption test and hemolysis test were negative, while cholesterol test, Meilan reduction reaction and tetrazolium nitrogen chloride reduction reaction were all positive. PCR products ofMycoplasma bovis showed the specific band of 1911 bp. Sequence analysis indicated that 16S rRNA of FJ-HJ showed 99.7% similarity with that ofMycoplasma bovis strain PG45. The results showed that the isolated strain was Mycoplasma bovis , proving the population ofMycoplasma bovis in Fujian Province. This study provides a basis for the prevention and control ofMycoplasma bovis in Fujian.
