  Screening of Zinc Enriched Rice Germplasm Resources in Heilongjiang Province 2017, 33 (20):  1-4.  DOI: 10.11924/j.issn.1000-6850.casb16070040 Abstract + ( 902 )   PDF ( 1091KB )   ( 364 ) To evaluate the zinc content of north rice materials comprehensively, 137 rice germplasms from Heilongjiang Province were analyzed using ICP- MS method in field experiment. The results showed that, under the same condition, the difference of rice germplasm zinc content from Heilongjiang Province was significant. The variation ranged from 10.92-25.47 mg/kg with the mean values of 16.15 mg/kg and the ratio of the maximum and minimum was 2.33. The result of frequency analysis of 137 rice germplasms zinc content showed unimodal normal distribution. The difference analysis was done in 4 accumulated temperature zones of Heilongjiang Province, the results of zinc content mean value patterns showed that zone 3＞zone 4＞zone 2＞ zone 1. L6, L170 with high level of zinc content were selected. These works will provide theoretical basis for reasonable utilization of local germplasm and new variety breeding in Heilongjiang Province.

  Analysis of Genetic Diversity and Identification of Maintainer Line for K-CMS Germplasm Resources in Wheat Using SRAP Markers 2017, 33 (20):  5-10.  DOI: 10.11924/j.issn.1000-6850.casb16070044 Abstract + ( 902 )   PDF ( 1797KB )   ( 377 ) To analyze the genetic background of the maintainer line for K-type cytoplasmic male sterility (CMS) in wheat, screen the excellent parents for breeding, and improve hybrid breeding efficiency in wheat, the genetic diversity of 30 maintainer lines for K-CMS germplasm resources of wheat were assessed using SRAP markers in this study. SRAP markers were used to amplify the polymorphic loci in the 30 lines, the genetic distances between cultivars were calculated by using Nei and Li methods. In 2 years field experiment, the average of phenotypic traits about plant height, panicle length, spikelet number, spike length, spike grass weight, grain number and grain weight per spike, length of flag leaf and so on were carried out in normal standard, using Euclidean distance to calculate the genetic distance among varieties, and comparative analysis was conducted. Analysis showed that 82 pairs of SRAP primers revealed polymorphism, the polymorphism rate was 68.3% , 208 polymorphic bands were obtained, the average of each pair of primers produced 3.68 polymorphic bands, showed high polymorphism. The results of cluster analysis based on molecular markers and pedigree analysis results were consistent. Taking the total average genetic distance as the scale, the results of cluster graph could be divided into 6 categories. Mantel detection showed that there was a very significant correlation between the genetic distance matrix of SRAP marker data and the expression pattern of the genetic distance matrix (r =0.8123,t =11.325>t 0.01). SRAP marker is an effective method to detect the genetic difference among cultivars, which can provide theoretical basis for the research of genetic difference of germplasm resources of K-CMS in wheat. Studies also confirm that the genetic difference between a backbone parent and the breed derived from it is generally small.

  Effects of Nitrogen Application Method on Yield and Quality of Strong Gluten Wheat 2017, 33 (20):  11-15.  DOI: 10.11924/j.issn.1000-6850.casb16120103 Abstract + ( 643 )   PDF ( 1243KB )   ( 281 ) The study aims to improve the cultivation technology system of strong gluten wheat. The strong gluten wheat‘Zhou yuan9369’was used as test material, the effects of nitrogen application amount, topdressing proportion, topdressing time on yield and quality of strong gluten wheat were studied. The results showed that the yield and crude protein content increased with the increasing of nitrogen application amount within a certain limit. The yield and nitrogen application amount showed quadratic curve relationship, and 300 kg/hm2 was the best nitrogen application amount in this experiment. The yield increased firstly with the decreasing of topdressing proportion, and then decreased. The crude protein content decreased with the decreasing of topdressing proportion, 3:7 was the most appropriate topdressing proportion in this experiment. Different topdressing time had different effects on grain yield and quality of strong gluten wheat, topdressing at jointing stage was the best in this experiment. The optimal plan of this experiment was nitrogen application amount 300 kg/hm2, topdressing proportion 3:7, and topdressing at jointing stage.

  High Yield Cultivation of Coix in Plateau Region —A Case Study of Shizong County in Yunnan 2017, 33 (20):  16-22.  DOI: 10.11924/j.issn.1000-6850.casb16090126 Abstract + ( 768 )   PDF ( 1173KB )   ( 379 ) To explore the yield potential of Coix in plateau area of Yunnan Province, and select the best Coix variety, planting density and fertilizing quantity in Shizong County, the authors carried out an orthogonal experiment with three factors and four levels, according to varieties (lines), planting density, fertilizing quantity. The results showed that the variety of‘Shiyi I’had the highest yield potential in the plateau of Shizong County. Moreover, the highest yield for all varieties (lines) occurred under the combination of low density (83370- 100050 plants/hm2) and moderate amount of fertilizing quantity (300- 450 kg/hm2). The treatment A1B3C3 ‘( Shiyi I’, 100050 plants/hm2, fertilizing quantity 450 kg/hm2) was the optimal combination model of variety, planting density and fertilizing quantity, and the yield reached 7143.60 kg/hm2. The optimal combination model can be recommended in Shizong County of Yunnan Province, and other regions with similar environment.

To improve water use efficiency and yield by means of the competition and complementary between intercropping crops is a difficult point of water use in intercropping. By literature reviews, the authors systematically analyzed the research achievements of water use efficiency of intercropping crops and its evaluation, and then explored the research difficulties and research priorities. The results could be applied to improving agricultural water use efficiency, and providing a theoretical basis for the sustainable development of agriculture.

  Cell Penetrating Peptides Mediated Transformation of Triticale Microspore 2017, 33 (20):  29-34.  DOI: 10.11924/j.issn.1000-6850.casb16070132 Abstract + ( 611 )   PDF ( 2152KB )   ( 264 ) In this study, the microspores from Triticale c.v. AC Ultima were used as explant to study the feasibility of cell penetrating peptides (CPPs) mediated transformation to a plant single cell. The microspores at mid-to-late-uninucleate stage were isolated and cultured in liquid suspension midium. The mexture of cell penetrating peptides (CPPs) and the plasmid DNA only with GFP gene’s expression frame were introduced into microspores. The transformants were screened by PCR and the expression of foreign GFP gene was identfied with fluorescence stereoscope. 21 transformed plants were regenerated, and 10 transformants were homozygous doubled haploid (DH) plants among them. The analysis of PCR confirmed 3 DH lines with positive results in T1 progeny, and GFP fluorescence could also be observed in the immature embryo of these lines. All the results indicated that exogenous GFP gene was well integrated into the genome of Triticale and inherited to offspring. This paper provides an effctive and feasible approch of CPPs mediated transformation, which will be applied in genetic transformation of plant single cell.

  Cloning and Expression Analysis of mtACP3 Genes in Peanut 2017, 33 (20):  35-42.  DOI: 10.11924/j.issn.1000-6850.casb16100110 Abstract + ( 608 )   PDF ( 1693KB )   ( 481 ) To investigate the mechanism of acyl carrier protein (ACPs) in the pathway of plant fatty acid synthesis, one ACP gene (AhmtACP3) was isolated from peanut by traditional PCR. The expression characteristics of the gene were analyzed by quantitative real- time RT- PCR (qRT- PCR). The length of AhmtACP3 gene was 859 bp. The length of open reading flame (ORF) was 390 bp, and its genomic sequence was 543 bp with 2 exons and 1 intron. It encoded a protein of 129 deduced amino acid residues. The molecular weight was 14.5345 kD and its isoelectric point was 5.22. The deduced protein was predicted to locate in mitochondria. Phylogenetic analysis indicated that peanut mitochondrial ACPs fell into two clusters. AhmtACP1 was grouped with AhmtACP2, which was distinct from AhmtACP3. qRT- PCR indicated that AhmtACP3 transcripts were more abundant in flowers than in any other tissues tested, followed by cotyledons and roots. The expression levels ofAhmtACP3 were the highest at the initial stage of seed development, but gradually decreased in abundance during the later stages.

  Active Ingredients Determination of Astragalus Aqueous Extract and Their Influence on Mycelium Growth of Five Edible Fungi 牛 宇,张丽珍,,,, and 2017, 33 (20):  43-49.  DOI: 10.11924/j.issn.1000-6850.casb16120029 Abstract + ( 759 )   PDF ( 1346KB )   ( 397 ) To investigate the contents of main constituents in the aqueous extract of Astragalus and their effect on the mycelium growth of Pleurotus ostreatus, Lentinus edodes, Auricularia auricula, Pleurotus nebrodensis and Pleurotus eryngii, the contents of total saponins, total flavonoids, total phenolic acids and total polysaccharides in the aqueous extract of Astragalus were determined using ultraviolet spectrophotometry, and the contents of calycosin-7-O-β -D-glucoside and fomononetin-7-O-glucoside in the extract were analyzed using high performance liquid chromatography. The results showed that the aqueous extract of Astragalus contained calycosin- 7- O-β - D- glucoside and fomononetin- 7- O- glucoside. The content of total saponins, total flavonoids, total phenolic acids and total polysaccharides in the extract was 62.67 mg/g, 56.48 mg/g,12.19 mg/g and 564.26 mg/g, respectively. Within the concentration range of test, the mycelium growth of Lentinus edodes was suppressed by Astragalus aqueous extract. For Pleurotus ostreatus, Auricularia auricula, Pleurotus nebrodensis and Pleurotus eryngii, the mycelium growth was promoted when the concentration of the extract was lower than certain thresholds, and was obviously inhibited when the concentration of the extract was above those thresholds. Pleurotus eryngii showed the highest mycelium growth inhibition threshold value, it was 1.5 g/100 mL. The main constituents of the aqueous extract of Astragalus were polysaccharides, saponins and flavonoids. At low concentrations, the extract promoted the mycelium growth of the tested edible fungi, whereas at high concentrations, it inhibited the mycelium growth.

  Purity Identification of Chinese Cabbage Hybrid‘Fuchun No.1’by SRAP Markers 2017, 33 (20):  50-54.  DOI: 10.11924/j.issn.1000-6850.casb16070095 Abstract + ( 721 )   PDF ( 1532KB )   ( 338 ) ‘Fuchun No.1’is a new late bolting spring Chinese cabbage cultivar breeded by Fuzhou Institute of Vegetable Science. In order to identify the purity of‘Fuchun No.1’, 80 pairs of SRAP primers were used to amplify the DNA templates of‘Fuchun No.1’and its parental pooling. The pairs of primers which contained bands of parental complementary characteristics were screened out and used to identify the purity of‘Fuchun No.1’seedlings cultivated in field. In the study, the pairs of primer M2＋E4 which contained bands of parental complementary characteristics were screened out and used to distinguish parents and hybrids effectively. The pairs of SRAP primer were adopted to identify the purity of 102‘Fuchun No.1’seedlings and the seed purity was 94.12% . The identification of SRAP markers is consistent with field survey results. SRAP molecular markers can be used to identify the purity of‘Fuchun No.1’.

  Flower Characters Segregation of F1 Generation of Reciprocal Cross from Phalaenopsis‘Big Chili’and P.‘Red Swan’ 2017, 33 (20):  55-60.  DOI: 10.11924/j.issn.1000-6850.casb17040116 Abstract + ( 955 )   PDF ( 2165KB )   ( 369 ) To study the flower characters inheritance rule of red flower Phalaenopsis, and solve the problem of fungoid disease and late florescence in common commercial variety P.‘Big chili’, the hybrids were obtained from the reciprocal cross from P.‘Big chili’and P.‘Red swan’. Results showed that distinct segregation appeared in the F1 generation of reciprocal cross. The depth of red was in gradient change, and the segregation regular of reciprocal cross was the same. The flower color was controlled by several pairs of genes, mainly dominant genes, and the more the dominant genes existed, the deeper red the flowers were. Moreover, the flower heart color, the decorative pattern and leaf characters might be dominant heredity. The 3 characters of F1 generation of‘Big chili’and‘Red swan’were more like thos e of‘Red swan’, which could improve the leaf resistance of‘Big chili’to reduce the fungoid disease, and the rate of fungoid disease caused by honeydew secreting on leaves in F1 generation could be below 25%. The average value of flower diameter, flower number and inflorescence length were all below the parents, and the mid- parent and positive heterosis were not obvious.

  Calcium Treatment on Fruit Calcium Ion Distribution and Fruit Quality Establishment in Peach 2017, 33 (20):  61-65.  DOI: 10.11924/j.issn.1000-6850.casb16070037 Abstract + ( 762 )   PDF ( 1418KB )   ( 321 ) To address the effect of calcium element on peach fruit and keeping quality, and establish the optimum leaf spraying concentration, we investigate the calcium distribution in fruit flesh, leaf chlorophyll content, and its effect on SSC, weight, size, flesh firmness and keeping quality of fruit. The results indicate the content of Ca2 in flesh increase with the spraying concentration of calcium. The concentration of Ca2 in flesh is positive consistent to the change of spraying level. The Chlorophyll content is up to the highest lever when spraying 0.5% CaCl2, while the SSC, fruit size, fruit firmness and storage quality are up to the maximum when spraying 1.0% CaCl2. In addition Spraying 1.5% CaCl2 is benefit to the fruit firmness after storage for 3 days. In the Present study, we established the appropriate spraying concentration of CaCl2, and the result will provide information for investigating the effect of Ca2 on fruit quality.

  Influence of Nitrogen Fertilizer Combined Application in Different Soil Layers on Growth Development and Nitrogen Use of Summer Maize 2017, 33 (20):  66-70.  DOI: 10.11924/j.issn.1000-6850.casb16110078 Abstract + ( 799 )   PDF ( 1216KB )   ( 329 ) The study aims to understand the effect of nitrogen management in different soil layers on growing development and nitrogen use of summer maize, using improved summer precision seeder with subsoiling and layered fertilizer. Two nitrogen fertilizer types, two soil depths with two nitrogen fertilization ratios were set. The effects of nitrogen management in different soil layers on leaf area index, dry matter accumulation per plant, yield and nitrogen partial factor productivity of summer maize were studied. The results indicated that the leaf area index, dry matter accumulation, yield and nitrogen partial factor productivity of summer maize with subsoiling and layered fertilization technology all obtained significant increase, compared to traditional fertilization technology with the same nitrogen amount. The yield and nitrogen partial factor productivity were increased by 5.5%-7.9%. Of the different subsoiling and layered fertilization technologies, the application of compound fertilizer with topdressing urea at bell stage obtained greater dry matter accumulation, yield and nitrogen use efficiency. While, it had no significant difference with controlled release fertilizer applied as base. The subsoiling and layered fertilization with compound fertilizer applied at 8cm and 12cm soil layer with ratios of 3:7 obtained the highest yield and nitrogen partial factor productivity both increased by 7.9%, which was the best model for subsioling and layered fertilization technology. Therefore, in summer maize areas of direct-seeding on wheat stubbles, the subsoiling and layered fertilization technology should be applied to improving yield and nitrogen efficiency.

  Effect of Liquid Organic Fertilizer on Photosynthetic Characteristics and Yield Formation of Cotton Under Drip Irrigation 徐海东,董合林,,徐文修,,, and 2017, 33 (20):  71-77.  DOI: 10.11924/j.issn.1000-6850.casb16090116 Abstract + ( 684 )   PDF ( 1344KB )   ( 377 ) To optimize fertilization technology of cotton under drip irrigation and study the effects of liquid organic fertilizer on growth and development of cotton, the authors adopted a single-factor randomized block test design, and studied the effects of liquid organic fertilizer application in different growth periods on photosynthetic characteristics, dry matter accumulation and yield of cotton. The results showed that (1) liquid organic fertilizer application could promote plant growth and development, increase photosynthetic rate and yield; with the increase of application rate of liquid organic fertilizer,LAI andSPAD value of all treatments increased and then decreased, and reached the maximum at boll stage, the maximum value in seedling stage and bud stage were 3.25 and 62.63 of liquid fertilizer C; (2) net photosynthetic rate (Pn ), transpiration rate (Tr )、stomatal conductance (Gs ) reached the highest under treatment C, while the intercellular CO2 concentration (Ci ) was the lowest (188.67 μmol/mol) under treatment C; (3) the dry matter accumulation during the whole growth period, bolls in pre-midsummer, bolls in midsummer, bolls of per plant and quality all reached the best under treatment C and reached significant difference level (P <0.05) compared with other treatments, the yield of treatment C was significantly higher than that of others by 27.01%-42.97%. Suitable liquid organic fertilizer application time improvedLAI , chlorophyll content and photosynthetic efficiency, and laid a good foundation for the dry matter accumulation and yield formation.

  Effects of Sowing Dates and Nitrogen Rates on Growth and Yield of Rape New Strain 159GP10 王瑞平 2017, 33 (20):  78-82.  DOI: 10.11924/j.issn.1000-6850.casb16070062 Abstract + ( 619 )   PDF ( 1217KB )   ( 360 ) Under the conditions of field experiments, new rape (named 159GP10) was conducted which included different sowing dates (September 30th, October 10th, October 20th and October 30th in 2015) and nitrogen (N) fertilizer treatment ( 0, 90, 180, 240 and 360 kg/hm2), and the main agronomic characters and grain yield were analyzed. The result shows that: there exits a great difference in growth process of 159GP10 at all sowing dates, but they do not change after the initial flowering. Meanwhile, the rape growth process was also affected under the different N fertilizer treatment, which indicates that rape in N(0 )treatment would be maturity earlily and its was earlier than 360 kg/hm2 by 8 days. With the increasing of N fertilizer, the number of green leaves, maximum leaf length and leaf width were gradually increasing trend, but the plant root diameter under 180 kg/hm2 was the highest. With sowing date postponing and N fertilizer reduction, strains height, effective number of branches, main sequence length and pods of rape plant are decreased, especially, pods was reduced significantly (p<0.05). Under different sowing date treatment, yield of October 10th treatment was the highest(3315.84 kg/hm2), that shows October 10th> September 30th > October 20th > October 30th.Under different N fertilizer treatments, the yield of 180 kg /hm2 was the highest (3157.18 kg/hm2).

  Heavy Metal Pollution of Agricultural Products: Present Situation in Some Areas of Yunnan 2017, 33 (20):  83-89.  DOI: 10.11924/j.issn.1000-6850.casb16070064 Abstract + ( 808 )   PDF ( 1251KB )   ( 691 ) The quality and safety of agricultural products is a common concern issues in the world and a agriculture development problem which our country attaches great importance. To understand the agricultural products’ heavy metal content in part of Yunnan and evaluate its edible security, the research of heavy metal pollution in parts of Yunnan provinces’ (mainly for the potentially contaminated area) main agricultural products were analyzed. Found that in parts of yunnan some metal tail mine pose a metal pollution hazard, heavy metal content exceed to security restrictions, crops, vegetables, economic crops, wild fungus resources and so on in the farming had certain security risks of heavy metals, Heavy metals content in Chinese traditional medicine, tea, tobacco is relatively safe. Based on the analysis of cause which agricultural products’ heavy metal content exceeds in yunnan province, the paper put forward the suggestions and countermeasures to solve the pollution of heavy metals in yunnan. The local government and relevant enterprises should improve the awareness of heavy metal pollution, strengthen supervision and management of planting industry, form a complete system of heavy metal pollution, and ensure the quality safety of agricultural products.

  Comparison of Two Short-time Heavy Precipitation Processes from Doppler Weather Radar Products 2017, 33 (20):  90-94.  DOI: 10.11924/j.issn.1000-6850.casb16070043 Abstract + ( 652 )   PDF ( 1668KB )   ( 284 ) To improve the forecasting and early-warming ability of severe convection weather in Jingzhou, and enhance meteorological service for agricultural production, the authors analyzed two short- time strong precipitation processes occurred on May 15 and June 1, 2015, by using Doppler radar data in Jingzhou. The results showed that: (1) the maximum radar echo intensity of these two processes were more than 50 dBz andstable, and“train effect”occurred during the moving of radar echo; (2) mesoscale convergence occurred in both processes, and the location of strong echo was consistent with the location of mesoscale convergence in velocity field; (3) most of the time, the cloud base height of these two processes were 2-3 km, and the cloud top height were around 12 km; obvious jump of echo top height and vertically integrated liquid (VIL) existed before the generation of storm; the VIL increments of the two processes were 10 kg/m2 and 20 kg/m2 and the increments of echo top height were 3 km and 9 km, respectively; (4) the vertical integrated liquid water contents were relatively stable in the storm development phase, were 40- 50 kg/m2 and 30- 40 kg/m2, respectively; the echo mass center height of the two processes were 4-5 km, and the maximum reflectivity height were 3-6 km at most of the time except June 1, when the sinking movement occurred; (5) downburst probably occurred on June 1 according to comprehensive analysis of the information of storm tracking.

  A Strong Hail Weather: Radar Characteristics and Its Impact on Agriculture in Changzhi 郭璐 and 裴坤宁 2017, 33 (20):  95-102.  DOI: 10.11924/j.issn.1000-6850.casb16110095 Abstract + ( 730 )   PDF ( 2252KB )   ( 494 ) To find the cause of hail and its impact on agriculture, the authors used Doppler radar reflectivity, radial velocity, vertical accumulation of liquid water content (VIL) products, combining with high and low air circulation situation and ground data, to analyze a strong hail weather process on June 13, 2016 in Changzhi. The results showed that: (1) the hail weather was made by the combined action of high altitude cold trough and warm low pressure; (2) strong echo and three body strong scattering (TBSS) had good indicative significance on early warning of hail; convergence and divergence of velocity could judge echo for the future development trend and enhance forecasting accuracy; when there was a certain continuous TBSS, TBSS could be used as the auxiliary index of strong hail warning; VIL was one of the effective tools for identifying hail and had a good role in forecasting the hail; (3) strengthening the hail monitoring, to realize“surface”warning by timely monitoring “points”was an effective supplementary means to reduce the hail damage to agricultural production.

  Cross-sectional Areas of Muscle Fibers in Different Parts of Xinjiang Brown Cattle and Local Yellow Cattle 2017, 33 (20):  103-106.  DOI: 10.11924/j.issn.1000-6850.casb16080018 Abstract + ( 887 )   PDF ( 1238KB )   ( 497 ) The study aims to compare the differences of muscle fiber area in different parts of Xinjiang brown cattle and local yellow cattle, and provide a basis for the studying the formation of meat quality of Xinjiang brown cattle. Three 2.5-3 year-old and healthy Xinjiang brown cattle and local yellow cattle under the same feeding conditions were selected to conduct the experiment, respectively. Muscle tissue samples were collected in tenderloin, silverside, sinew meat, chuck tender, topside, highrib, rump, cucumber, brisket, ribeye, etc. And the techniques of frozen tissue section, HE staining and Motic Image Advanced 3.2 software were used to measure the cross-sectional area of muscle fiber. The results showed that the average cross-sectional area of muscle fiber of topside and cucumber in Xinjiang brown cattle were significantly smaller than that in local yellow cattle (P <0.05). However, the average cross-sectional area of muscle fiber in sinew meat, tenderloin, highrib, rump and brisket in Xinjiang brown cattle were significantly bigger than that in local yellow cattle (P < 0.05). There was no difference in the average cross-sectional area of muscle fibers in other parts between Xinjiang brown cattle and local yellow cattle. In conclusion, cross-sectional areas of muscle fibers had diverse differences in different parts between Xinjiang brown cattle and local yellow cattle.

  Isolation and Identification of Mycoplasma bovis FJ-HJ Strain 2017, 33 (20):  107-112.  DOI: 10.11924/j.issn.1000-6850.casb16070088 Abstract + ( 613 )   PDF ( 1358KB )   ( 347 ) One strain of Mycoplasma bovis was isolated from lung tissue of an infected beef cattle characterized by gasp and runny noses in Fujian Province. To identify this strain, colonial morphological observation, biochemical test, specific polymerase chain reaction (PCR), cloning and sequencing of 16S rRNA gene were conducted. The results showed that colonies of this strain, which was named as FJ-HJ, were like fried eggs with brown protrusion in the center. FJ- HJ could not ferment glucose, not hydrolyze arginine, and not decompose urea. Hemadsorption test and hemolysis test were negative, while cholesterol test, Meilan reduction reaction and tetrazolium nitrogen chloride reduction reaction were all positive. PCR products ofMycoplasma bovis showed the specific band of 1911 bp. Sequence analysis indicated that 16S rRNA of FJ-HJ showed 99.7% similarity with that ofMycoplasma bovis strain PG45. The results showed that the isolated strain was Mycoplasma bovis , proving the population ofMycoplasma bovis in Fujian Province. This study provides a basis for the prevention and control ofMycoplasma bovis in Fujian.

  Genetic Effect and Detection of SNPs in Candidate Genes Related to Milk Production Traits of Xinjiang Brown Cattle 2017, 33 (20):  113-123.  DOI: 10.11924/j.issn.1000-6850.casb16080102 Abstract + ( 546 )   PDF ( 1372KB )   ( 440 ) This study aimed to explore the association between polymorphisms of candidate genes related to milk production traits and somatic cell score (SCS), and explore the genetic feature of polymorphism from these SNPs in Xinjiang brown cattle. Based on DNA pools with the Re-sequencing technology to gain SNPs data, totally screened 17 SNPs of FASN, FSHR, HAL, GPIHBP1, NIBPL, C9, PDE9A, PTK2, STAT1, ZNF66, ZNF567, ZNF7, ZNF623 and ZNF804A genes, at the same time, combining with Sequenom MassArray Genotype technology to detect the genetic polymorphism. The genetic effect of 17 SNPs on 305 d milk yield, milk fat percentage, milk protein percentage, milk lactin percentage, total solids content and SCS at Xinjiang Urumqi farm, Cattle farm in Tacheng area and Xinjiang Tianshan Animal Husbandry Biological Engineering Co., Ltd. were analyzed by GLM procedure of SAS8.1.Totally 262 Xinjiang Brown cattle. The results showed that the 17 SNPs were in Hardy-Weinberg equilibrium, and 9 SNPs had significant (P<0.005) or extremely significant (P<0.001) association with milk production traits, and these SNPs were located in FASN, FSHR, HAL, ZNF66, ZNF7, ZNF804A, GPIHBP1 and PDE9A genes. Association analysis results of this study provide a theoretical basis and reference for the molecular marker-assisted selection in Xinjiang brown cattle.

  Expression of β-catenin in Uterus of Artificially Induced Oestrous Sheep 2017, 33 (20):  124-127.  DOI: 10.11924/j.issn.1000-6850.casb16070069 Abstract + ( 567 )   PDF ( 1302KB )   ( 317 ) To investigate the changes in the expression of β-catenin in uterus of oestrous sheep which was artificially induced, expression changes of β-catenin on the 10th, 12th, 14th, 16th and 18thday after sheep oestrus were detected respectively by Real-Time PCR and semi-quantitative PCR. Real-time quantitative and semi-quantitative PCR results showed that: two detection methods could detect mRNA expression in sheep uterus in different periods of β-catenin, and in 10th to 18th days were shown to decrease after increasing trend. The transcriptional level of β-catenin on the 10th day was significantly higher than that of the other days (P<0.05). In addition, β-catenin mRNA expression on the 12th day reduced significantly (P<0.05), and then remained relatively constantly, but significantly increased on the 18th day (P<0.05). Real-time PCR and semi-quantitative PCR can be used to detect the relative expression level of sheep uterine tissue of β-catenin mRNA; during the 10th to 18th day after sheep oestrus, the transcriptional level of β-catenin presented a decrease-increase trend.

Using the methods including literature reviewing, expert consultation and seminars, the research on division and composition of professional dimension was carried out in depth in the frame structure of agricultural engineering construction standard system. In addition, detailed statements of standards in the Agricultural engineering construction standard system were ascertained by the above methods. At the final, the Agricultural engineering construction standard system was built in China. The study lay a foundation for agricultural engineering construction standard system revision.

  Application Status of New Media in Chinese Agricultural Kernel Periodicals 2017, 33 (20):  138-143.  DOI: 10.11924/j.issn.1000-6850.casb16080103 Abstract + ( 710 )   PDF ( 1319KB )   ( 353 ) The analysis of application status of new media in Chinese agricultural kernel periodicals can provide the guideline for the development strategy of agricultural technical periodicals under the new media environment. The data of the application of new media in agricultural kernel periodicals was analyzed and the impact factor of above mentioned periodicals were derived from China Journal Citation Reports in 2015. The feature of dispersion was presented in the application of new media in Chinese agricultural kernel periodicals in general. Meanwhile, the difference of application of new media between different discipline existed. In the same category, the impact factor of agricultural kernel periodicals with application of new media was higher than that of one without application of new media. The application of new media techniques would broaden the present research on the application of new media technique and provide support for the development of strategy research of the application of new media technique. The application of new media techniques could improve the influence of agricultural kernel periodicals and become main means for increasing influence of periodicals in future.

  Efficacy of Tilapia Culture Based on Recirculating Aquaculture System 2017, 33 (20):  144-148.  DOI: 10.11924/j.issn.1000-6850.casb16100027 Abstract + ( 609 )   PDF ( 1291KB )   ( 749 ) A high-density aquaculture system was built for experiment with tilapia as the culture object for three months in this study, in order to study its regulated ability on water quality and fish growth. The system was constructed by a vertical precipitator, drum microfiltration machine, moving-bed biofilm reactor, multiple chamber spraying oxygenator, trickle degassing tower and sterilization devices, and had high performance in water treatment. The results showed that the feeding and growth of tilapia were normal under the high-density aquaculture system. The stocking density increased from 3.5 kg/m3 to 32 kg/m3 rapidly with the food coefficient of 1.72. The system had significant regulated ability on water quality, which was stable after treatment. When the amount of daily water exchange was maintained at about 5%, the temperature was maintained at 23-28.0℃, concentrations of ammonia nitrogen, nitrite nitrogen, and dissolved oxygen were 0.272, 0.067-0.21 and 7.40 mg/L, respectively, and COD was 7.15 mg/L. The results prove that applying this recirculating system in tilapia culture is feasible and efficient.

In recent years, PBDEs have been detected in a variety of environmental media and human bodies around the world. PBDEs pollution near the e-waste dismantling area, sewage outfall and large industrial zone is the most serious. Having the characteristics of biological accumulation, environmental persistence and biological toxicity, PBDEs cause the environmental and health problems which are widely concerned. Along the two chains of water - sediment - edible animal - human and atmosphere - human, we reviewed the recent pollution status of PBDEs in environmental media and human, pollution sources, bioaccumulation characteristics and exposure scenarios of PBDEs in diet and daily life of human. By comparing the pollution level of PBDEs in China and foreign countries, we revealed problems at the present and proposed corresponding countermeasures, which would provide a reference for further research, such as environmental toxicology and ecological risk assessment, of PBDEs in China,.
