Abstract: The paper aims to evaluate main components and antioxidant activity of Platycodon grandiflorum extracts. Water and ethanol extracts were prepared by reflux extraction, polysaccharide was prepared by water extracting- alcohol precipitating method. Platycodin D content was detected by HPLC method, spectrophotometry was used to detect the content of total flavonoids and polysaccharide. The antioxidant activities of the extracts were detected based on the clearance of hydroxyl radical and DPPH · model. The content of total flavonoids in ethanol extracts from Anhui, Jilin and Shaanxi was 1.35%, 1.30% and 1.08%, respectively. The clearance rate for scavenging DPPH· and ·OH of ethanol extracts from Anhui and Jilin was higher than that from Shaanxi. The content of polysaccharide in water extracts from Anhui, Jilin and Shaanxi was 0.45%, 0.39% and 0.35%, respectively. The clearance rate of polysaccharide for scavenging DPPH· and ·OH from Anhui was the highest, that from Jilin was the next, and that from Shaanxi was the lowest. The clearance rate of P. grandiflorum ethanol extracts for scavenging DPPH· and ·OH was higher than that of water extracts and polysaccharide. Ethanol extracts have good in vitro antioxidant properties and provide some reference for the development of related antioxidant products.
