Cloning and Bioinformatics Analysis of Sucrose Transporter Gene Glyma18g15950 in Vegetable Soybean

Abstract: In order to reveal the characteristics of vegetable soybean sucrose transporter in the molecular level, we homologously cloned the sucrose transporter gene Glyma18g15950 from the vegetable soybean variety “Mindou 6”. After sequencing, it was found that the length of CDS sequence was 1794 bp, encoding a polypeptide containing 597 amino acids. Bioinformatics analysis showed that the isoelectric point (pI) and molecular weight (Mw) was 5.81 and 64.30 kD, respectively, and this protein is a hydrophobic protein. The secondary structure of amino acid residues were also predicted by SOMPA, and the results showed 42.47% for α-helix, 13.71% for β-sheet, 39.63% for random coil and 4.18% for β-turn. Based to the amino acid sequence and structural analysis, Glyma18g15950 protein contained one conserved domain (MFS_2 domain). SignalP analysis showed that the protein has no signal peptide. Phylogenetic tree analysis showed that the Glyma18g15950 protein had the closest genetic relationship with Glycine soja and the homology was 97.34%. The alignment between this protein and 9 SUC proteins from Arabidopsis thaliana showed that Glyma18g15950 was closely to AtSUC3 and belonged to the SUT2 subfamily.
