Response Surface: Optimizing the Medium for Gliotoxin Produced by Trichoderma viride

Abstract: Response surface method was used to optimize the culture medium for gliotoxin production by Trichoderma viride. Evaluation of the effects of the 6 factors relating to the basic medium was carried out through Min Run Res IV design, which revealed the significant influence factors. Steepest ascent design(SAD) and central composite design(CCD) were applied to determine the midpoint and the optimal value respectively, and the optimized culture medium was obtained. Glucose, soymeal, KH2PO4 and ZnSO4 were the most significant influence factors screened by Min Run Res IV. Response surface analysis of the SAD and CCD indicated that the optimal culture medium was composed of 26.54 g/L glucose,10 g/L yeast powder,15.72 g/L soymeal, 0.452g/L KH2PO4, 0.0582g/L ZnSO4 and 0.1g/L FeSO4, which resulted in a 59.71% increase of the titer of gliotoxin compared with the original medium.
