Proanthocyanidin Extraction Process from Brown Cotton Fiber: Optimized by Response Surface Method

Abstract:

To improve the yield of proanthocyanidin from brown cotton fiber, and optimize the extraction process, the brown cotton fibers were used as materials to study the effect of methanol concentration, solid-liquid ratio, ultrasonic time and temperature on the extraction efficiency of proanthocyanidin, and to obtain the optimized extraction conditions. Proanthocyanidin was extracted by ultrasonic combined with methanol-n-butanol hydrochloride, the extraction process was optimized by response surface methodology. The results showed that the optimum parameters were methanol concentration of 83%, solid-liquid ratio of 1:60, ultrasonic temperature of 42℃, ultrasonic time of 18 min, and the extraction rate was (7.16±0.03)%. The extraction yield of proanthocyanidin by the optimized method was improved by about 4% compared with that of the general method. The extraction process optimized by response surface methodology has high extraction efficiency, and the experimental operation is simple and fast, which can be used to extract proanthocyanidin in batches. The study lays a foundation for the research on biological activities and the development of products of proanthocyanidin.
