Establishment of the Explant Regeneration System for Genetic Transformation and Transformation by Agrobacterium rhizogene of Siraitia grosvenorii(Swingle)C. Jeffrey

Abstract: Stem segments and leafy discs from in vitro grown plantlets of Siraitia grosvenorii(Swingle)C. Jeffrey cv.Qingpinguo were used as explants,the regeneration system of recipient material for genetic transformation was developped. Transformation mediated by Agrobacterium rhizogene was also explored in this experiment. The results showed that the regeneration ability of stem was better than leafy and stem segments were sensitive to Agrobacterium . Stem segments were good recipent material for gene transformation. On the MS basic medium with 1 mg/ LBA, 1 mg/ LKT, 0.2 mg/ L NAA and 30 mg/L sugarose, the frequency of bud induction of 20-day-old stem segments was 100%, every stem segments produced 9.7 buds in average and the survival rate of transferred plantlets was over 90%. Injury to explants improved the number of induced bud. Hairy roots were formed from stem segments infected by Agrobacterium rhizogene. The induction and the culture of hairy root of Siraitia grosvenorii(Swingle)C. Jeffrey will be helpful for the study and development of its medicinal functions.
