Comparative Study on the Biological Characteristics of Protoplast Fusion Strains and Parent of Phellinus baum ii

Abstract:

Abstract: OBJECTIVE To select high biomass , extrapolysaccharide and total flavones-yielding protoplast fusion strains of phellinus baumii. METHODS The esterase isozyme of five strains were examined by polyacrylamide slab gel electrophoresis.And the selected metabolic parameters in submerged cultures of five strains were determined.RESULTS The results shown that there were two steady identical basic isozyme bands among strains, and the three selected protoplast fusion strains had there peculiar pattern bands. The biomass of strainSR002 and SR003 was three times as strainSA04,and strainSR002’s extrapolysaccharide content had increased 1.8 fold compared with strainSA02. StrainSR005 had the highest phytoesterase activity. strainSR002 and SR003’s relations were very near.CONCLUSION The three selected protoplast fusion strains have a high potential for producing extrapolysaccharide and mycelium in submerged culture after further testing of genetic stability.
