The Sequencing and Analysis of Amino Acids in Silkworm Pupa

Abstract:

The aim was to develop new resources about silkworm chrysalis, 13 amino acids were determined in silkworm pupa. The contents of amino acids were analyzed by column derivatization with OPA, reversed phase high performance liquid chromatography (RP-HPLC), gradient elution method and fluorescence detection. The contents of aspartic acid, glutamic acid, serine, histidine, glycine, threonine, alanine, taurine, methionine, valine, phenylalanine, isoleucine and leucine contents were 63.97 mg/g, 72.48 mg/g, 32.46 mg/g, 52.21 mg/g, 6.531 mg/g, 35.63 mg/g, 31.37 mg/g, 21.39 mg/g, 1.806 mg/g, 71.01 mg/g, 68.08 mg/g, 35.86 mg/g and 25.34 mg/g, respectively. Total amino acid content was 518.15 mg/g with 45.88% essential amino acids. RP-HPLC method for determining amino acids is simple, rapid, sensitive and accurate.
