Study on Technical of Reducing Tissue Culture Browning of Amorphophallus Konjac Bulbs

Abstract:

Using Amorphophallus konjac bulbs as explants, the research studied method of reducing browning of explants in tissue culture process. By using MS+6-BA 1 mg/L+ NAA 1 mg/L as the basic medium and the orthogonal experimental method to study the impact on browning with different agar concentrations, adsorbent and light, dark two kinds of different culture conditions. The results showed that the callus growth speed was enhanced and the browning of explants was under control in the combination of MS+ 6-BA 1 mg/L+ NAA 1 mg/L+ sugar 30 g/L+ agar 6 g/L+AC 0.5 g/L+PVP 0.05 g/L. It can make to reduce the browning rate of 38.3%, callus induction rate increased to 78.3%. The konjac bulbs explants the browning can be effectively controlled, which add a certain amount of activated carbon (AC) and polyvinylpyrrolidone (PVP) in the culture medium and in the dark conditions, its callus can be promptly formed.
