The Effect of Salicylic Acid on Coumarin Content in Fraxinus Hybrid Cell

Abstract:

In order to study the effects of SA on the contents of coumarins in the suspension cultured cells of Fraxinus Hybrid which paved the way for establishing plant cell suspension cultures of Fraxinus Hybrid, three different concentration of SA (25, 50, 100 mg/L) were applied in the suspension cultured cells which had been cultured for 96 h. Then the cell growth and content of coumarins were determined at 8 h, 24 h, 48 h, 72 h and 96 h after treatment. The results showed that the signal molecule SA raised the contents and productions of coumarin and fraxinol to different respectively. Among the three concentrations, 100 mg/L of SA brought the best effect to coumarins accumulation. The content and production of fraxinol in the suspension cultured cells which was harvested at 72 h after treatment were 0.5704 mg/g and 0.2595 mg which were 114.59 % and 35.03% higher than control, respectively. The variation trends of the content and production of coumarin were the same; they reached the maximum at 72 h after treatment and were 0.2470 mg/g and 0.1124 mg which were 112.39% and 56.56% higher than the control. It concluded that the appropriate concentrations of SA could effectively promote the accumulation of coumarins in Fraxinus interspecific hybrid suspension cell.
