Abstract:
RSM in extraction process optimization for yield of chlorogenic acid (CGA) in callus of Apocynum venetum, the results laid the foundation for further exploration of extracting CGA in the callus on large scale. The yield of CGA was taken as the index, CGA yields in the callus were determined with HPLC and compared, the optimum extraction process was determined. On the basis of single factor analysis test, combined with central composite experimental design principle, response surface methodology (RSM) was used to optimize extraction process. The optimum extraction method from callus of A. venetum was microwave-assisted extraction. On the basis of single factor analysis test, experiments were carried out and combined with RSM, the optimum microwave-assisted extraction process was as follows: extraction time 40min, extraction power 40w, ratio of solid to liquid 1:40, the CGA maximum yield reached 0.4911 mg/g(D.W), interaction between extraction time and extraction power had a significant impact. Establishment of the two polynomial mathematical models had the important role for CGA extraction from callus of A. venetum.
