Correlative Analysis between Polymorphism of PGC-1α Gene and Meat Quality Traits in Qingyuan Partridge Chickens

Abstract:

[Objective] The PGC-1α gene is the key gene impacting meat quality . In order to find out a mutation site having more influence on meat quality, this article studied the polymorphism of PGC-1α gene, which can provide the foundation of the next functional verification of protein. [Method] We found the polymorphism site of PGC-1α gene of Qingyuan Partridge Chickens by PCR-SSCP and PCR-LDR. Fat content, shear force and WLR(Water lost rate of breast muscle)tested as meat quality indicators were grouped according to different genotypes. And then we analyzed the impact of different genotype on meat quality via analysis of variance and multiple comparison. [Result] We screened two polymorphism sites SNP1 and SNP2 in Qingyuan Partridge Chickens, which are both mutated from G to A. SNP1 is in an intron, which has no effect on meat quality. SNP2 is a sense mutation in the exon 8 with a significant effect on WHC (Water Holding Capacity). Hens’ meat quality is better than cocks’, and there is extremely significant difference between the two sexes as to the shear forces. As for SNP2, the meat quality of AG genotype is better than of AA and GG genotypes.
