Changes of the Transcription Levels of Molecules Associated with Endogenous Antigen Processing and Presentation of Porcine Skin-derived Dendritic Cells Infected with PCV2 in vivo

Abstract:

Healthy Large White piglets of forty-day-old were euthanized on 3, 7, 14, 21 and 35 days post-infected with porcine circovirus type 2 (PCV2) and their skin-derived dendritic cells (DCs) were collected for analyzing the transcription levels of molecules (LMP7, UBP, MHC-I, calreticulin) associated with endogenous antigen processing and presentation by real-time fluorescent quantitative PCR (real-time FQ-PCR). The results showed that the transcription levels of LMP7 mRNAs were decreased significantly on 3 days post inoculation (DPI) (P < 0.05). UBP and calreticulin mRNAs were consistently higher during the experiment, only with significant differences on 21 and 35 DPI (P < 0.05) for UBP. And MHCⅠwere decreased significantly on 7 DPI (P < 0.05). All of the above results indicated a reduction in the ability of endogenous antigen processing and presentation of porcine skin-derived DCs during the early period of PCV2-infected piglets.
