Study of Factors Affecting on Physical Germination of the Apple Pollen

Abstract:

This study used four kinds of pollen (‘Hongxing’, ‘Meihong’, ‘Hehong’, ‘Quanjiahong’) as the test material to determine the pollen vitality of all varieties by two ways including staining (TTC and peroxidase method) and germination in vitro. Pollen germination and growth experiments in vitro were observed under different culture conditions. The results showed that: in staining and germination experiments, there was an obvious diversity among the activity between pollen vitality, the ‘Hongxing’ was the highest, the ‘Quanjiahong’ was the lowest. The germination and elongation of pollen first increased and then decreased with culture temperature and concentration increasing. The most optimum concentrations of pollen germination and growth was different under the different culture temperatures, high-concentration can inhibit the pollen’s germination and tube growth. There were different culture condition between the highest rate of pollen germination and the maximum length of the pollen tube, and the optimum temperature and concentration of the growth of pollen tube were higher than the optimum conditions pollen germination because of the vigorous metabolism.
