In Vitro Rapid Multiplication and Propagation from Leaves of P. simonii Carr.×P. deltoides cv. ‘Shanhaiguanensis’

Abstract:

In order to develop new technology for Populus simonii gene preservation and enhance its resistance to disease and pests, leaves of hybrid clone of P. simonii Carr.×P. deltoides cv. ‘Shanhaiguanensis’ were used as explants to study it’s in vitro culture and leaf regeneration system, and to explore the effect of different prescriptions of hormone on induction, differentiation of adventitious bud, and rooting. The results showed that: the optimum medium for adventitious buds regeneration of P. simonii Carr.×P. deltoides cv.‘Shanhaiguanensis’ was MS+0.1 mg/L 6-BA+0.04 mg/L NAA+0.005 mg/L TDZ with the maximum adventitious bud frequency (90%), number of shoots (6.8), shoot length (2.14 cm). The optimum medium for rooting of shoots was 1/2 MS + 0.1 mg/L IBA with a rooting efficiency of 90%. The plantlets thus obtained were successfully hardened and transplanted to greenhouse. 10 mg/L kanamycin could inhibit the bud regeneration of “P. simonii Carr.×P. deltoides cv. ‘Shanhaiguanensis’”, 20 mg/L kanamycin could inhibit the root regeneration of P. simonii Carr.×P. deltoides cv. ‘Shanhaiguanensis’ from the shoots. The results of this study will not only contribute to the pest-resistant area of P. simonii and the related studies of genetic transformation, but also contribute to provide important technical support for the preservation of gene pool of P. simonii.
