Abstract:
Partial sequence of the rp operon of phytoplasma associated with mulberry yellow dwarf disease (MYD) was amplified by Polymerase Chain Reaction (PCR) employing primer pair rpF/rpR, sequenced and analyzed by corresponding biology software. The results show that a 1240bp DNA fragment in the rp operon of phytoplasma was amplified from the infected mulberry(GenBank accession No. GQ373255).The fragment includes partial sequence of rps19 gene, complete sequence of rpl22 and rps3 genes. The sequence shares 96.6-99.9% identity to those of other representative phytoplasmas in the 16SrⅠ- rp subgroup. Phylogenetic tree shows that the MYD phytoplasma and mulberry dwarf (MD) phytoplasma are clustered into the same subgroup, 16SrⅠ- rp-B subgroup.
