Tissue Culture and Rapid Propagation of Raspberry (Rubus idaeus) Cultivar ‘Kivigold’

Abstract:

The sterilization time of HgCl2 (0.1%) in initial culture, suitable type and concentration of phytohormone in proliferation and rooting culture media of adventitious bud, suitable matrix in transplant were selected using the stem segment with an axillary bud of raspberry (Rubus idaeus) cultivar ‘Kivigold’ as explants, and also suitable sugar during proliferation culture process was studied. The results showed that in initial culture, the optimal sterilization time of 0.1% HgCl2 was 10 min, the explants can grow well on MS medium with only 6-BA; The single factor experiment revealed that cytokinin mainly affects on proliferation while auxin affects on growth during proliferation process of adventitious bud, and medium with 6-BA concentration below 1.5 mg/L and NAA concentration below 0.5mg/L was more appropriate; Sucrose is more beneficial to bud growth during proliferation process of adventitious bud; Through a further combinative selection, it was determined that the medium suitable for proliferation of ‘Kivigold’ adventitious bud was MS + 1.00 mg/L 6-BA + 0.10 mg/L NAA (containing 20 g/L sucrose and 5.9 g/L agar, pH 5.8), the optimum medium for rooting of ‘Kivigold’ adventitious bud was 1/2 MS + 0.10 mg/L NAA (containing 20 g/L sucrose and 5.9 g/L agar, pH 5.8); Peat soil + perlite at the rate of 1:1 was the suitable matrix for transplanting, the survival rate was 93.33%.
