Study on the sequencing and phylogenetics analysis of the rice chloroplast DNA petG-trnP fragment

Abstract:

The rice chloroplast petG-trnP fragment was selected to directly sequence and analyze in this study. The results demonstrated that the length of the rice petG-trnP intergenic spacer was very conservative and 316 bp with 242 bp non-coding region except the trnW sequence. The non-coding region was separated into two parts with 116bp and 126bp respectively. The G+C content was relatively low and about 37.19%-38.02% in the non-coding region. The 5 rice materials Clustal W analysis showed that the petG-trnP fragment structure was variable, there were 7 variable sites (2.89%) in the non-coding region. As a result, this indicted that it contained a few phylogenetic information and would provide the direct molecular evidence to clarify the Oryza species origin and evolution.
