Screening、Identification and Degradation Conditions of Cellulose Decomposing Bacteria Bacillus pumilus T-7

Abstract:

In order to get one strain which can degrade the cellulose, more than 41 strains were isolated from vegetable field, animal manure and silage feed by heating gathering spore bacteria strains and Congo red culture medium transparent hydrolysis circle, and tested their transparent of fermentation liquor and identification to cellulose through secondary screening. The T-7 strain has the highest capability, its enzyme activity was 1678.89U/mL. The mycelium shape and the bacterial colony, physiological and biochemistry experiments and determination of the sequence of 16S rDNA of the fungi had been done. The strain was identified as Bacillus pumilus. The degradation condition and the best cultivation condition were studied. It showed that the concentration of inoculum 1 billion viable cells / 1Kg corn stalks, add the secondary carbon and nitrogen sources in the 2%sugar and 2% urea case to the corn, the degradation rate of cellulose was 40.34%. The results were explored a new resources for the biodegradation of cellulose ,and laid foundations for the large-scale production and use of the straw.
