Lodicules Microscopic Observation of Noncleistogamy and Cleistogamy Wheat

Abstract:

Wheat (Triticum aestivum L.) is a self-pollinating crop. But most of wheat is noncleistogamy. Contrasting with noncleistogamy, cleistogamy can keep the purity of pure line varieties, reduce the risk of spike disease infection and insure the seed set under stress environment. In order to find the reason which lead to the noncleistogamy and cleistogamy in Wheat (Triticum aestivum L.) and lay the foundation for the exploration of the mechanism in noncleistogamy and cleistogamy. In this study the noncleistogamy variety Luomai 4 and cleistogamy variety Huapei 3 were used as materials. The lodicules were taken while the materials were heading. The depths of lodicules were compared through frozen section, its depths were measured as well. The lodicules’ depths in Luomai 4 were 1.67 times the lodicules’ depths in Huapei 3. The whole clearing method was also taken to discover highs, widths and cell sizes difference between two varieties. The observation was carried out by the differential interference microscope. It was found that the lodicules’ highs and widths between two types (noncleistogamy and cleistogamy) were not difference. The lodicules’ cell sizes of two types were also no significantly difference.
