    Construction of Tremella fuciformis Expression Vector and Expression of EGFP Gene

    Yeast-like conidia of Tremella fuciformis were used as the material. Plasmid pEGFP and pCAMBIA1300 were digested by Hind III and EcoRⅠ restriction enzymes and ligated to transform. We constructed Tremella fuciformis expressed vector pCB-BEGFP contained enhanced green fluorescent protein gene which was regulated by gpd promoter of Agaricus bisporus. Electroporation was performed to transfer plasmid DNA of pCB-BEGFP into yeast-like conidia from Tremella fuciformis. The chromosomal DNAs of the four transformants and the nontransgenic strain as the control were used as the templates for the PCR using the EGFP-specific primers. When the products were analyzed by the agarose gel electrophoresis, the expected 750bp amplified bands appeared. By fluorescence microscopy, distinct green fluorescence could be observed from the colonies of YLCs grown on RM agar plates and could not be observed from the colonies of YLCs which did not contain plasmid pCB-BEGFP. Expression of EGFP was detected by SDS-PAGE analysis of TCP of one transformants, as expected, a dark band of the protein was visualized at 27kD by Coomassie Brilliant Blue staining. These results indicated the EGFP gene had been integrated into the genome of transgenic Tremella fuciformis strains and was expressed successfully by regulation of gpd promoter of Agaricus bisporus.
    Effect on the Mycelium of Volvariella volvacea Under the Different Liquid Culture Conditions

    For the Volvariella volvacea V37 under the liquid culture industrialization production provides the concrete reference formula to the straw mushroom, and plants each kind of environmental condition request in the production process. The Volvariella volvacea V37 under the liquid culture conditions was studied, the different culture medium formula, the raise temperature, liquid volume in the bottled, the inoculation rate, pH and the culture time to its mycelium biomass were influenced. The results showed that: the optimal fermentation medium was formula B. Medium capacity 80 mL, inoculation rate 10%, pH was 6 to 9, the optimal culture conditions were temperature 30-32℃ and the culture time was from 6 days to 8 days.

    Studies on the Optimization of the Fermentation Conditions of Exopolysaccharide Produced by Lactobacillus casei KL1 from Tibetan Kefir

    In this paper, the Lactobacillus casei from Tibetan Kefir was used to screen for improving the environment factors of the yield of exopolysaccharide(EPS). The four factors affecting synthesize of exopolysaccharide of the fermentation conditions were determined by the four factors and three levels orthogonal experiments [L9(3)4]. The result was as follows: fermentation temperature—37℃, fermentation time—12h, origin pH of culture medium—6.5, amount of inoculation—5%. With the optimization conditions,the yield of exopolysaccharide was increased 1.39 times.
    Studies on Virus-free of Lentinula edodes

    with unique fragrances and a variety of health functions, Lentinula endodos is one of main cultivated mushroom in China. However, in some regions of China in recent years have witnessed varying degrees of mushroom virus disease,causing severe influences in yield and quality. Through experiences it showed that the virus is widely existed in Lentinula edodes.Three Lentinula edodes strains were selected to eliminate virus by picking up hypha tips(less than 0.2mm in length) . The results showed that:some strains with one band of dsRNA (11kb) left in Lentinula edodes strains were got, which called incomplete eliminated strain. only one strain was got with no dsRNA in it ,which called virus-free strain. the virus- elimination rate is very low by this method. The growth rate of the three kinds of strains in different pH were measured, it showed that the virus-free strain grew faster than virus-infected strain in pH 8 condition. cellulase, xylanase, laccase and polyphenol oxidase activity of the three kinds of strains were mensurated ,the result showed that the enzyme activity of Laccase and Polyphenol oxidase in virus-free strain were higher than that of virus-infected strain.Through the cultivation experiments ,the fruit body of virus-free strain is hypertrophy than others, and with a stronger anti-pollution ability.

    Studies on the Optimization of Fermentation Conditions of Bile Salt Hydrolase by Lactobacillus casei KL1 from Tibetan Kefir

    In this paper, the Lactobacillus casei of bile salt hydrolase screened from Tibetan Kefir were used to study environment factors of increasing enzyme vires. For four factors to affect synthesize of bile salt hydrolase, fermentation conditions of high yield bile salt hydrolase were determined by four factors and three levels orthogonal experiments [L9(3)4]. As follows: glucose 2%, Soya-peptone 2%,fermentation temperature 37℃, amount of inoculation 2%.Under optimization conditions,the vires of bile salt hydrolase was enhanced to 7.4 times.

    At present, it is noticeable for some mushroom-farms to use pesticides to control the damages caused by insects or disease in China. The incidents of pesticide residues in edible fungus products over Maximum Residues Limits at home and abroad have happened frequently. It seriously effected on the quality and safety of edible fungus products and it became a bottleneck which constrains edible fungus products export of china. In this paper, on the basis of a comprehensive analysis of occurring disciplinarian of the common pests and diseases in the progress of edible fungus planting, it is suggested that edible fungus integrated pest management(IPM) is recommended include edible fungus production technology management, fungus diseases and insect pests critical control point, the use of pesticides Standardization and quality control throughout the edible fungi safety production and others. It is offered some advices to absolutely ensure edible fungus production safety and quality.
    Primary Research about the Seeds Production of Iphigenia indica Knuth

    Iphigenia indica Knuth is a kind of small herbs in Iphigenia genus, Liliaceae. It is almost close to extinction. The bulb contained colchicines. The seeds production of the plant had been researched. The appropriate conditions for seeds germination were 25~28℃, pH6.2~7.5, 1800Lx illumination for 18 hours, hot water treatment at 50℃ for 12 to 18 hours, low temperature treatment(9℃)for 5 days, or variable temperature treatment(4℃ and 9℃)for 5 days, 180 days stored.
    Establishment of SSR-PCR Technological System and Its Preliminary Application in Genetic Diversity Analysis for Lentinula edodes

    Abstract: SSR primers of Lentinula edodes were developed using ISSR-suppression PCR method and Primer 3 program, and L16(45)orthogonal design was used to optimize main factors affecting SSR-PCR amplification system. The optimal SSR-PCR system was established as follows, within 20μl reaction system, there were 1.5 mmol/L Mg2+, 75 ng template DNA, 0.25 mmol/L dNTPs, 0.50 μmol/L primer and 1.5U Taq DNA polymerase. The optimal annealing temperature was determined as 62℃ by gradient PCR. Based on the optimal system, suitable results were obtained when five primer pairs were used to amplify genomic DNA of eight strains .Cluster analysis well reflected geographical original relation of these strains. The eight strains were classified into two groups using 0.5 similarity as a cut-off point. GroupⅠmainly consisted of strains from North China, while group Ⅱ comprised those from South China.

    Abstract : It was determined the virulence of Verticillium lecanii MZ041024 on Larvae and pupae of Lycoriella pleuroti. The bioassay results indicated that insecticidal activity of ethyl acetate was higher than that of others, their mortality rate of larvae and pupae respectively were 91.79% and 88.94%, with LC50 values of 1.745×105 spores /ml and 4.723×105 spores /ml ; LT50 was respectively3.51d and 3.78d, against Larvae and pupae of Lycoriella pleuroti , respectively.

    Studying the Techniques for Reuse of Spent Mushroom Substrates

    To face the problem of vast spent mushroom substrate (SMS) produced from mushroom industries, we analyzed the nutritive compounds of various SMS and studied the bio-efficiency of applying SMS as substitutes for sawdust, cottonseed hull and wheat bran on mushroom cultivation, as well as the effects on organic fertilizer for the yield and quality of Navel orange production. Results showed the possibility of re-utilizing SMS as substrates for Pleurotus cornucopiae, Tricholoma lobayense and Pleurotus abalonus with the bio-efficient for about 64.95%、84.65% and 70.49%, respectively, which were similar as it of CK group obtained. Comparing to CK group, use SMS as organic fertilizer increased the average output of Navel orange for 5.54kg per tree significantly, improved fruit quality and modified garden soil structure. Results indicated that re-utilization of SMS would apply an important role in recycling of natural resources in agricultural production.

    The different shape mushroom viruses are part of mycoviruses and most of them possess undivided dsRNA genomes, but some of them possess the positive single-strand genomes. The undivided dsRNA genomes composed of a coat protein (CP) gene and an RNA dependent RNA polymerase (RdRp) gene are the best feature of genomes of mushroom viruses. Mushroom viruses have been sequenced and the structure and function of their genomes have been analyzed. DsRNA technique and the method of enzymology make the study of the mushroom viruses easy to reach to molecular level. Especially dsRNA technique applied to the Pleurotus ostreatus. Mushroom viruses have special replicative form, which have two phases, and the theory of a cellular self-replicating mRNA as their origin of them is attractive because of their obvious ancient origin. These contents were reviewed in this paper. We expatiate on the further research progress in the relationship between mushroom viruses and their hosts. It is important to research further the mushroom viruses and to screen the free viruses strain.
    Studies on the Characteristics of Bile Salt Hydrolase Produced by Kluyveromyces marxianus M3 from Tibetan Kefir

    Abstract: To investigate the characteristics and dynamic principle of the bile salt hydrolase produced by Kluyveromyces marxianus M3 from Tibetan Kefir, the single factor and multi levels orthogonal experiments was applied. Enzyme activity was tested under the different contitions:pH4~8, temperature31~43℃, concentration of substrate 4~8mmol/L, and different chemical reagents(SDS、EDTA、carbamide、Cu2+、Mg2+、Ca2+、Fe3+、Al3+、Mn2+ )during 30 min reaction between bile salt hydrolase and substrate. The optimal reaction condition of bile salt hydrolase was pH6.0, concentration of substrate 7mmol/L, temperature37℃. Metal ion of Mn2+、Fe3+、Ca2+ had comparatively large elevation effect on the enzyme activity, and other chemical reagents had few effect on the enzyme activity. The kinetic constant Km of bile salt hydrolase was 2.60 mmol/L. Strain M3 entered the stationary phases after 18~21h. The enzyme activity reached the maxlmum during log phase growth at18h. The experiments results showed the category of dynamic principle was growth couple synthesis type.

    The Phylogenetic Relationship of Several Varieties for Export of Lentinula edodes and Comparative Experiment of Cultivation

    In order to select and breed new varieties of Lentinula edodes for export, the phylogenetic relationship of several strains of L. edodes was analyzed based on ribosomal DNA ITS sequences, and comparative experiments of cultivation between the two selected strains and some others cultivated in China were also investigated. The results indicated the moderate genetic diversity among these strains for export, and they could be clustered into two groups. The strains qh01, qh04, qh05 and qh06 were clustered into one group, which indicated a close phylogenetic relationship within the group. However, the single strain qh02 was clustered into another group, which inferred a far phylogenetic relationship with other strains. Comparative experiments showed various agronomic traits among the tested strains, and especially, the two strains qh02 and qh06, showed excellent agronomic traits, and consequently inferred that the two strains were suitable for export.

    This experiment studied on the edible amaranth, the Malabar spinach vegetable executed the nitrogen level to conduct the research differently to the vegetables nitric acid salt content influence. It’s for executes the nitrogenous fertilizer, the reduced fertilizer pollution provides the rationale as well as the promoted special vegetables. Carries on the design through the experimental statistics method to test and to analyze .The result indicated that, the NO3- content increases in certain scope along with the N application increases, and the fertilizer surpasses, the output can drop, while the nitric acid salt content still had the rise. At the same time, the vegetables in the different developmental stage, the weather situation and the applying fertilizer time length, the NO3- content also can differ from.

    The paper studies factors of the Extraction of Lentinan Polysaccharide by Microwave Treatment, the factores include ratio of material、extracting temperature、extracting time、microwave heating time and extracting times. The results showed the optimal conditions were: 25 of ratio of liquid to solid, 80℃ of temperature,3 hours of heating time, 30 min of microwave heating time, extraction once. The highest extraction ratio 7.70%.
    Effects of Bleeding Sap of Sponge Gourd as Fresh-keeping Agent on pleurotus citrinopileatus

    Pleurotus citrinopileatus were used to study the effects of bleeding sap of sponge gourd as fresh-keeping agent on Mushroom.The results showed that bleeding sap of sponge gourd could effectively prolong storage stage of mushroom and be conducive to maintaining commodity quality. Of all the treatments, 0.75 volume fraction of bleeding sap of ridged sponge gourd had the best fresh-keeping effect, compared with the control treatment, the emergence of different symptoms of deterioration such as pileus split,color change,decay and smell-up deferred 4~6days; when reserved at 10 0C for 12 days, weightlessness treated with 75% RSG bleeding sap was 54.17% of control treatment, and was lower than control and other treatments highly significantly.

    Throuth orthogonal test , the study covered the effects on the rate of cap opening , PPO activity and ADF by controlling concentration of NaCl, time of colour fixation and storage temperatures. The result indicated that with proper handing to treatment mushrooms could significantly reduce the cap opening and quality deterioration during storage. The experimented results was further analogged by using regression equation. The estimated values were found to be identical with the measured values. Compared with regular low-temperature storage, the mushrooms had better quality and physical-chemical index in low-temperature and controlled atmosphere storage.
