无核荔枝生长素反应因子基因克隆及序列分析

Cloning and Sequence Analysis of Auxin Response Factor of Seedless Litchi

摘要: 为获得在无核荔枝中胚珠退化过程中差异表达的生长素反应因子基因(ARF),对其全长的克隆并进行分析,根据构建的无核荔枝胚珠败育SSH消减文库的生长素反应因子EST序列,提取高质量的RNA,通过SMART-RACE技术,获得一个生长素反应因子3501 bp的核苷酸序列。应用生物信息学软件进行分析,预测该序列含有2550 bp的开放阅读框,推导的蛋白质为849个氨基酸,具有B3、Auxin_resp和AUX_IAA保守区与结构功能域。分析表明该基因为ARFs基因家族中的一个新成员,进化树上与芒果亲缘关系最近。

Abstract: To obtain the auxin response factor gene (ARF) that was differentially expressed in ovule degeneration of seedless litchi, in this study the gene was cloned and analyzed. The EST encoding ARF was obtained from young abortive ovule of seedless litchi via SSH (suppression subtractive hybridization), extracted high quality RNA and cloned the total cDNA sequence by SMAT-RACE method, bioinformatics analysis showed that it included an 2550 bp open reading frame which encoded a predicated protein with 849 amino acids, moreover, it had B3, Auxin_resp and AUX_IAA conserved region and functional domains, indicated that it was a new member of the ARFs gene family and got the closest relatives with Mangifera indica.
