南方根结线虫食道腺表达基因7E12影响植物的初步研究

摘要: 南方根结线虫(Meloidogyne incognita)是一种重要的植物寄生线虫,它的食道腺产生的分泌蛋白是对寄主造成危害的致病因子。鉴定这些编译分泌蛋白的基因的特性,是认知南方根结线虫对植物致病性或寄生性机理的关键所在。本研究通过使用烟草根部特异表达基因TobRB7的△0.3缺失的启动子,替换植物过表达载体pBI-121的 CMV(烟草花叶病毒) 35S 启动子,构建了南方根结线虫食道腺细胞表达的基因7E12的植物表达载体pBI-Trp-7E12和pBI-Trp。pBI-Trp在本研究中作为空白对照使用。利用农杆菌(Agrobacterium rhizogenes)侵染花粉管通道法把这两个载体转化了模式植物拟南芥。通过利用卡那霉素抗性培养基对已转化的拟南芥进行筛选,获得了T1代转基因植株。这为研究鉴定该基因的特性奠定了基础,以利于进一步研究南方根结线虫对植物的致病性机理。

Abstract: The south root-knot nematode (Meloidogyne incognita) was an important plant-parasitic nematode, the secretion proteins from its esophageal gland cells were the pathogenic factors which would hurt the host. Identification of the genes encoding these proteins was beneficial to know the pathogenicity and parasitism mechanism of the south root-knot nematode. The plant root specific expression vector pBI-Trp-7E12 of Meloidogyne incognita esophageal gland cell specific expression gene 7E12 and pBI-Trp were constructed by using the △0.3 truncated promoter of tobacco root specific expression gene TobRB7 to replace the CMV (cauliflower mosaic virus) 35S promoter of plant over-expression vector pBI121. In the research, pBI-Trp was used as blank control. These two vectors were transformed to the research model plant: Arabidopsis thaliana by using Agrobacterium rhizogenes via pollen tube channel infection method. Filtered for the transformated Arabidopsis thaliana by using the kanamycin resistance medium and obtained the T1 generation of transgenic plants. This study laid a foundation for the research and identification of the gene characterization of 7E12, so as to further the research on the pathogenic mechanism of south root-knot nematode to the host plants.
