3个大豆品种及其骨干亲本的亲缘关系和指纹图谱分析

Analysis of Genetic Relationship and DNA Fingerprinting of Three Soybean Cultivars and Their Core Parental Lines

摘要: 为了研究江苏淮北地区大豆推广品种及其骨干亲本的亲缘关系和指纹图谱,以3个淮豆系列品种及其8份骨干亲本品种为试验材料,利用CTAB法提取基因组DNA,采用SSR分子标记对11份大豆品种进行亲缘关系和指纹图谱分析。结果显示:利用筛选出的46对SSR引物在11个大豆品种中共检测出125个等位变异,每对引物可检测到2~5个等位变异。使用非加权类平均法进行聚类分析,大部分供试材料间的遗传变异较小,遗传相似系数为0.5781~0.9609。SSR标记遗传距离为0.0391~0.4219,平均0.2773。从上述引物中选取5个核心引物构建的指纹图谱能够鉴别3个淮豆系列品种。上述结果不仅用于品种鉴定及其知识产权保护,还为有效选配亲本拓宽遗传基础提供了理论基础和技术支持。

Abstract: In order to study genetic relationship and DNA fingerprinting of commercial soybean cultivars and their patental lines in Huaibei Region, Jiangsu, total genomic DNA of 3 high-yielding (Huaidou) soybean cultivars and their 8 core parental lines was extracted by using CTAB method. The genetic similarity coefficients and DNA fingerprinting of the above-mentioned materials were conducted by using SSR markers. 46 pairs of primers were selected and distributed evenly on the whole genome, of which 2 to 5 alleles were detected by each pair of primers. A total of 125 polymorphism fragments were amplified, whose average number was 2.7 per pair of primers. The cluster analysis was conducted by using unweighted pair-group method with arithmetic means (UPGMA). The genetic variation in most of the experimental materials was lower, and their genetic similarity coefficients changed from 0.5781 to 0.9609. The genetic distance between SSR markers were from 0.0391 to 0.4219 and their average value was 0.2773. The fingerprinting of 5 core primers selected from the above primers could be markedly distinguished between the 3 high-yielding (Huaidou) cultivars. These results not only can be used to detect the cultivars (lines) authenticity and protect intellectual property rights, but also to provide theoretical basis and technical support for effective selection of parents to broaden the genetic basis.
