大豆花粉活力测定方法的比较研究

Soybean Pollen Vitality Testing by Different Methods

摘要: 为研究不同方法测定大豆花粉活力的问题,选取‘绥农4’、‘吉林47’和‘CK-P’为研究对象,通过蔗糖、琼脂、硼酸、温度单因子正交试验筛选出的培养基进行离体萌发,比较I2-KI染色法、TTC染色法和离体萌发法3种花粉活力测定方法的不同。结果表明:I2-KI染色法容易操作,因这种方法较离体萌发法简单,测定结果比较准确,成本低,所需时间短,常用来观测大豆花粉活力。TTC法染色效果与I2-KI染色法接近,没有差异,也可作为检测大豆花粉活力的方法。离体萌发法测定大豆花粉活力的方法和很多因素有关,与I2-KI染色法和TTC染色法有显著差异,且其培养基的配制繁琐,所以用I2-KI染色法和TTC染色法来测定大豆的花粉活力。

Abstract: In order to study different methods for determination of soybean pollen vitality, SuiNong4、JiLin47 and CK-P were studied, through single factor orthogonal test to sucrose ,agar, H3BO3 and temperature, filtered the culture medium for pollen germination , the difference of pollen viability between I2-KI, TTC staining and in vitro germination were compared. The results showed that the I2-KI staining method was easy to operate, because the method was more simple than the in vitro germination method, the determination result was accurate, the cost was low and the time was short,so it was used to test pollen vitality . TTC and I2-KI staining method was similar, there was no difference, but also can be used as a method to detect soybean pollen vitality. The method of vitro germination of pollen viability was related to many factors,and I2-KI and TTC staining were significantly different, and its preparations were complex and cumbersome, so I2-KI and TTC staining method can be used to determine soybean pollen viability.
