茶饼病病原—Exobasidium vexans侵染茶树叶片过程的形态学观察

The Morphology Observation of Infection Process for the Pathogen Exobasidium vexans of Tea Blister Blight against Tea Leaf

摘要: 明确茶饼病病原在侵染过程中的形态特征,对于病害诊断及有效防控具有重要意义。本研究采用过碘酸-希夫(氏)和甲苯胺蓝染色分析Exobasidium vexans侵染茶叶组织和细胞的过程。同时,采用台盼蓝和伊文思蓝染色及透射电镜对茶饼病样品植物细胞活力、及病原菌生长进行观察。结果表明,组织染色及电镜发现在茶饼病2~3阶段有大量担孢子萌发和菌丝生长,茶树叶片细胞营养也逐渐消耗,担孢子和菌丝活力降低,茶树叶片细胞趋于死亡。本研究通过染色方法发现该病害在侵染各阶段的形态特征,通过透射电镜发现病原对海绵组织细胞的结构影响。

Abstract: To determine the morphological characteristics during infection process for the pathogen of Tea Blister Blight, this should facilitate the disease diagnosis and effective control. This study used the stains of Periodic Acid-Schiff and Toluidine Blue O to analyse the infection process of the pathogen against the tissues and cells in tea leaf. Meanwhile, Trypan blue and Evans blue were used to analyze the cell viability of the tea leaf, and the status of basidiospore and hypha. The histological stain and TEM found that an amount of spore germinated and hypha grown during 2-3th phase, and the nutrient of tea leaf cells was gradually consumed, then the viabilities of the basidiospore and hypha were also decreased, tea leaf cells ultimately tended to die. This study found the morphological characteristics during various infection processes by the methods of the histological stain, and found the influence of cells from the spongy tissue triggered by the pathogen.
