BBD-response Surface Methodology Optimizes the Cultivation Conditions of Producing Gliotoxin by Trichoderma viride

摘要:

旨在优化绿色木霉产胶霉菌素培养条件。实验取绿色木霉产胶霉菌素培养条件中的5个重要影响因子,设计5因素3水平的响应面分析实验,数据用Design-Expert 8.0分析得出预测模型,验证模型的可靠性后,得出绿色木霉产胶霉菌素最佳的培养条件为:培养温度29.42℃、培养基初始pH 6.35、接种量5.44%、培养时间54.5 h、菌龄44.5 h,且在最佳培养条件下胶霉菌素效价较优化前提高了15.08%。

Abstract:

The aim is to optimize the cultivation conditions of producing gliotoxin by Trichoderma viride. 5 important factors in the gliotoxin cultivation from Trichoderma viride were chosen, and the response surface analysis experiment (five factors three levels) was designed. A predictive model was established through the analysis of the data by Design-Expert 8.0, and the reliability of the model was verified. The optimal condition of gliotoxin production was determined as follows: the cultivation temperature was 29.42℃, the initial pH value of the medium was 6.35, the inoculation volume was 5.4%, the cultivation was for 54.5 h, the bacterium age was 44.5 h. The titer of gliotoxin was increased by 15.08% compared with that before the optimization.
