鸡NYGGF4同源基因的克隆与表达分析

Chicken NYGGF4 Homologous Gene Cloning and Expression Analysis

摘要:

小鼠和人的NYGGF4基因参与前脂肪细胞的增殖调控,并可能与肥胖的发生发展有关。本研究首次克隆获得了鸡的NYGGF4同源基因的全长编码序列,利用生物信息学方法对鸡NYGGF4同源基因的遗传进化性和蛋白功能进行预测分析,并利用半定量RT-PCR技术对雏鸡的NYGGF4同源基因进行组织表达谱分析。结果表明:鸡NYGGF4同源基因的编码序列全长654bp,进化保守,在多个组织脏器中具有转录表达活性,编码蛋白具有一个磷酸酪氨酸作用结构域(PID),而编码PID结构域的蛋白广泛参与神经发育、免疫、组织平衡和细胞生长等生理过程,这表明鸡NYGGF4同源基因是一个可能具有多种功能的新基因。

Abstract:

Mouse and human NYGGF4 is a novel gene, which involves in regulation of adipose precursor cells proliferation and may be related to occurrence and development of obesity. In this study, full-length coding sequence of chicken NYGGF4 homologous gene was gained for the first time. Genetic homology and protein functions of chicken NYGGF4 homologous gene were predicted using bioinformatics methods, and tissue expression pattern was also analyzed using Semi-quantitative RT-PCR technology. The results show that chicken NYGGF4 homologous gene full-length coding sequence is 654nt, and evolution is conservative. In chicken, the gene expresses in a variety of organizations and possesses a phosphotyrosine interacting domain(PID).Encoding PID proteins widely involve in physiological processes, such as neural development, immune, organizational balance and cell growth, which indicates chicken NYGGF4 homologous gene is a novel gene that maybe possesses multiple functions.
