大白菜‘津育60’遗传转化前后组织培养条件的优化

The Screening of Tissue Culture Medium for Brassica compestris L. ssp. pekinensis Olsson cv. ‘Jinyu60’

摘要:

为大白菜的遗传转化研究建立较高频率的组织再生体系,选择白菜子叶—子叶柄作为外植体,通过正交试验优化了农杆菌侵染前后的组织培养条件。结果显示:侵染前的最佳分化培养基组合为MS培养基+3 mg/L 6-BA+0.3 mg/L NAA+4 mg/L AgNO3+2%蔗糖+0.8%琼脂,侵染后的最佳分化培养基组合为MS培养基+3 mg/L 6-BA+0.01 mg/L NAA+4 mg/L AgNO3+2%蔗糖+0.8%琼脂。6-BA与NAA的浓度比由侵染前的3:0.3调整为3:0.01。

Abstract:

To study the genetic transformation of Chinese cabbage-pe-tsai (Brassica compestris L. ssp. pekinensis Olsson ‘Jinyu60’), its tissue culture system was established. Cotyledons with petiole were used as explants, and the culture conditions were optimized by an orthogonal design. The results showed that, before and after genetic transformation, the optimum organization culture medium combination for ‘Jinyu60’ was: MS+3 mg/L 6-BA+0.3 mg/L NAA+4 mg/L AgNO3+2% sucrose+0.8% agar and MS+3 mg/L 6-BA+0.01 mg/L NAA+4 mg/L AgNO3+2% sucrose+0.8% agar, respectively. The concentration rate between 6-BA and NAA was 3:0.01.
