白灵菇原生质体制备条件的优化

Optimizing Conditions of Preparing Pleurotus nebrodensiss Protoplast

摘要:

以白灵菇单核菌丝为材料,利用溶壁酶进行原生质体的制备,考察各因素对原生质体产量的影响,采用单因素试验和正交试验确定最佳条件。结果表明:原生质体产量最高的条件是取菌龄为7天的液体静置培养的菌丝体,以0.6 mol/L甘露醇为稳渗剂,在酶解温度27℃、酶浓度2%的条件下酶解150 min。研究结果可为白灵菇原生质体融合及转基因研究提供理论依据。

Abstract:

The optimal conditions for the lywallzyme hydrolysis of the cultured monocaryon mycelia of Pleurotus nebrodensiss for protoplast production were investigate by single factor and orthogonal array design methods. The results showed that the highest production rate of the protoplast was emerged under the conditions that 7 days old liquid mycelium in the enzyme solution of 2% lywallzyme digested for 150 min at temperature of 27℃ with 0.6 mol/L mannitol as osmotic pressure stabilizer. Theoretical base for studying the protoplast fusion and transgenic of Pleurotus nebrodensiss was provided.
