果蝇卵巢管经固定和染色后的荧光效果研究

The Study on Fluorescent Features of Drosophila Ovarioles after Fixation and Staining

摘要:

为进一步研究不同生物材料的荧光染色特性,开辟荧光组织学的新领域,以黑腹果蝇卵巢为实验材料,解剖取出雌果蝇的卵巢管,通过不固定直接压片、用多聚甲醛固定液固定后压片及固定后再用不同染料染色后压片这3种方法处理后,分别在明场(白光)、绿色激发光、蓝色激发光和紫外激发光下观察、拍照。发现果蝇的卵巢管具有弱的自发荧光性,多聚甲醛可赋予卵巢管明显的荧光特性,固定后的标本经不同染料染色后荧光性分别出现不同的显著变化。通过综合分析认为,常规固定剂及染色剂可以分别赋予果蝇卵巢管中的生殖细胞和体细胞各自不同的荧光特性,也可赋予细胞内不同物质成份以各自不同的荧光特性。不同固定、染色剂赋予生物组织细胞新荧光特性的研究在组织学中将会有重要而广阔的应用前景。

Abstract:

In order to provide a trial exploration for studying the fluorescent features of different biological materials and opening up new areas of fluorescent histology, Drosophila melanogaste ovaries were used as experimental materials. The ovarioles were firstly dissected, and then treated in three ways: directly squashed material, squashed material after fixation with paraformaldehyde, and squashed material stained with five different deys after paraformaldehyde fixation. The squashes were observed and photographed under fluorescent microscope in bright field, and fluorescent fields after excitation with green, blue and UV light, respectively. The results showed that Drosophila ovarioles possesses weak autofluorescence, paraformaldehyde fixation endowed the material evident fluorescent features, and different staining endowed the material various fluorescent characteristics. Based on the above results, the following conclusion can be reached: fixatives and dyes can endow Drosophila ovarioles and different cell types, or even different cellular components specific fluorescent characteristics. Fluorescent techniques might open a bright applying field for the traditional histology.
