茶树腋芽离体培养中的褐化控制研究

Prevent browning of axillary buds in vitro culture of Camellia sinensis

摘要:

为降低茶树离体培养过程中外植体褐化程度,以‘凫早2号’当年生未木质化的春季健壮新梢为材料,带茎段和叶柄的单个腋芽为外植体,接种前用2.0 g/L PVP溶液浸泡1 h,采用正交试验研究不同培养基、起始培养温度和时间对外植体的褐化控制效果。结果表明,腋芽在1/4MS+BA 2.0 mg/L+IBA 0.1 mg/L+蔗糖20g/L+琼脂7.5 g/L培养基中,10℃下避光培养48 h褐变率最低,仅为5.6%;培养时间和培养基类型2个因素对降低腋芽褐化没有显著影响,温度对降低腋芽褐化有显著影响;温度因素外植体对褐化影响最大,培养基次之,培养时间最小,说明低起始培养温度是降低外植体褐化的主要因素。

Abstract:

The aim of this study was to evaluate the effects of strengths of MS medium, different culture temperatures and time on prevent explant browning in plants of Camellia sinensis cv Fuzao 2#. Axillary buds reserved a part of stalk excised from juvenile twigs origin of current year growth tea plants is explant. Explants cultured on basal medium supplemented with BA 2.0 mg?L-1 +IBA 0.1mg?L-1+sucrose 20g?L-1+agar 7.5g?L-1 after adsorbing phenolics used 2.0g?L-1 PVP solution for 1h. Minimum rate of explant browning (5.6%) were developed on 1/4MS at 10℃ under 48h dark conditions. The results showed no differences between strengths of MS medium and culture time in the percentage of explant browning, however, culture temperature have significant effect on explant browning. the effect of the factors on explant browning was in sequence of culture temperature>strengths of MS medium>culture time, low culture temperature is an essential factor in prevent explant browning. Successful reduces axillary buds browning in vitro culture by low temperature suggested that this strategy was viable for preventing explant browning.
