多胺及稀土对欧石楠诱导分化的影响

The Effect of Polyamines and Rare-earth Elements on Differentiation of the Bottles of Seedlings of the Erica carnea

摘要: 为探讨多胺(ployamines, PAs)及稀土元素(rare-earth elements)在植物组织培养应用中的理论基础和实践意义,以欧石楠继代瓶苗为外植体,采用组织培养的方法,研究不同浓度的多胺及硝酸铈和硝酸镧对其继代瓶苗诱导分化的作用。在基本培养基(1/2 WPM+ZT 1.0 mg/L+IBA 0.3 mg/L)中添加1.0 mg/L的亚精胺(Spd),欧石楠的平均分化倍数、平均分化苗高及胚性愈伤组织诱导率分别达到3.89、3.11 cm、98.30%;在基本培养基中添加1.0 mg/L的硝酸铈[Ce(NO3)3],欧石楠的平均分化倍数、平均分化苗高及胚性愈伤组织诱导率分别为6.9、4.0 cm、98.70%;在基本培养基中添加0.5 mg/L的硝酸镧[La(NO3)3],欧石楠的平均分化倍数、平均分化苗高及愈伤组织诱导率分别为5.5、3.89 cm、100%,这3组试验,显著性方差分析及多重比较结果显示,该处理相对于其他处理均达到极显著水平。低浓度的多胺及稀土元素对欧石楠继代瓶苗的分化及生长具有促进作用,而高浓度则起抑制作用。

Abstract: The induction and differentiation effects of various concentrations of ployamines, cerium nitrate and lanthanum nitrate on Erica carnea’s subculture test-tube seedlings were studied to provide theoretical basis and experimental basis of ployamines and rare-earth elements application in plant tissue culture. The method applied to plant tissue culture, regarded Erica carnea’s subculture test-tube seedlings as an explant, were discussed the effect of variable concentration of ployamines, Ce(NO3)3 and La(NO3)3 on the introduction of the subculture test-tube seedlings. The results showed that the average differentiation times, the average height of differentiation plantlet and embryogenic callus induction frequency of Erica carnea on basic medium (1/2 WPM+ZT 1.0 mg/L+IBA 0.3 mg/L) supplemented with 1.0 mg/L spermdine were 3.89, 3.11 cm and 98.30% respectively, which were significantly better than other treatment demonstrated by analysis of variance and multiple comparisons results; basic medium with 1.0 mg/L cerium nitrate showed the average differentiation times 6.9, the average height of differentiation plantlet 4.0 cm, and embryogenic callus induction frequency 98.70%, which were significantly better than other treatment demonstrated by analysis of variance and multiple comparisons results. Meanwhile basic medium with 0.5 mg/L lanthanum nitrate gave the highest embryogenic callus induction frequency 100%, the average differentiation times 5.5, the average height of differentiation plantlet 3.89 cm, which were also significantly better than other treatment indicated by analysis of variance and multiple comparisons results. The low concentration of ployamines and rare-earth elements acted as a stimulus to the differentiation as well as growth of Erica carnea’s subculture test-tube seedlings, otherwise, the high concentration caused the inhibition of the differentiation and growth of Erica carnea’s subculture test-tube seedlings.
