低温胁迫对香椿幼苗蛋白质含量及特异蛋白表达的影响

The Effect of Low-temperature Stress upon Protein Content and Component in Leaves of Toona Sinensis Seedlings

摘要: 以抗寒性强弱不同的河南西峡、江苏南京和福建霞浦3个种源1a生香椿为材料,研究了不同低温处理条件下可溶性蛋白质含量及组分的变化。结果表明,低温胁迫可以引起香椿叶片蛋白质含量的变化,但不同种源在不同温度下的表现不一致,同一种源在不同胁迫时间下的动态变化也不一致。SDS-PAGE电泳结果显示,抗寒性较强的河南西峡、江苏南京种源变化规律相似,在-5℃低温胁迫1~4d过程中,可溶性蛋白质含量呈现“升高-降低-再升高”的变化趋势,即胁迫1d后蛋白质即大量表达,谱带颜色变深,随后在第2、3天含量逐 渐下降,谱带变浅,第4天含量又开始增加。两种源分别诱导表达出分子量为27.6 KD、22.5 KD的特异蛋白。抗寒性较弱的福建霞浦种源可溶性蛋白质含量呈逐渐增加的趋势,但没有诱导出新的蛋白质谱带。各种源SDS-PAGE电泳结果显示的蛋白质谱带颜色的深浅变化与蛋白质含量变化基本一致。

Abstract:

Abstract: Under low-temperature stress, the changes of protein content and component in leaves of annual Toona Sinensis provenances seedlings, Xixia of Henan, Nanjing of Jiangsu, Xiapu of Fujian, were studied. The results indicated that the low-temperature stress could change the content and component in leaves of Toona Sinensis provenances seedlings. Different provenances had different changes under different temperatures. So did the same provenance under different stress period. SDS-PAGE electrophoresis indicated that low-temperature stress could induce specific proteins’ expression. The protein contents of Xixia of Henan and Nanjing of Jiangsu provenances, whose cold resistance were stronger, tended to change as ‘high-low-high’ pattern during one to four days under -5℃. Proteins were induced highly in the first day, and the band became deep. During the second and the third days, it’s the opposite. While in the forth day, it began to express more again. And there were two specific proteins expressed, the molecular weights were 27.6 KD and 22.5 KD for the two provenances respectively. The protein content of Xiapu of Fujian, whose cold resistance were weaker, tended to increase gradually, and had no new proteins appeared. By electrophoresis of SDS-PAGE, the changing of the color of the protein bands changed almost the same with the protein contents.
