黄瓜果实横径的遗传分析及分子标记

GENETIC ANALYSIS AND MOLECULAR LOCALIZATION OF THE FRUIT DIAMETER IN CUCUMBER

摘要:

本文选用果实横径粗的品系D07171、D0899和果实横径细的品系D0435-3-1、D0438-3按照Griffing完全双列杂交法Ⅳ配置成6个杂交组合,选取杂交组合D0435-3-1×D07171经自交构建F2群体,对黄瓜果实横径进行遗传分析SSR及分子标记。结果表明:黄瓜果实横径大小是由多基因控制的数量性状。果实横径符合AD模型,以加性效应为主,显性效应较小;广义遗传力和狭义遗传力均较高,分别为60.64%和57.26%。采用116对SSR引物对果实横径性状进行分子标记,最终检测到1个与黄瓜果实横径相关的QTL,位于CSWCT25-CSWCT29-CSWTA03连锁群上,距离引物CSWTA03较近,为1.98cM,对应的LOD值为3.22,可解释为7.19%的遗传变异。

Abstract:

In this study, selected big fruit diameter of D07171 and D0899, small fruit diameter D0435-3-1 and D0438-3, in accordance with the method of complete diallel crosses have been the preparation of 6 combinations to Analyze the Genetic model of fruit diameter. The combination of D0435-3-1×D07171 was selected as the largest different combination to get the F2 generation for test of normality, using SSR molecular marker technology to build genetic linkage map. Cucumber fruit diameter of many genes are controlled by the quantity of genetic. Cucumber fruit diameter in line with the additive-dominance (AD) genetic model for the additive effect of the main, dominant effect is less than additive effect. The broad-sense heritability and narrow sense heritability of the cucumber fruit diameter are both high, 60.64% and 57.26% respectively. In this study, 116 pairs of SSR primers in the DNA of two parents and between the pool filter, detected with a cucumber fruit diameter a QTL related sites, located in the genetic map of linkage group CSWCT25-CSWCT29-CSWTA03, and CSWTA03 closer distance for the 1.98 cM , the corresponding LOD value of 3.22, 7.19% explain the genetic variation.
