Near-isogenic Lines (NILs) Selection of Quality Protein Maize (QPM) By Marker Assisted Selection

Abstract: In order to enrich quality protein maize (QPM) new germplasms, some common germplasms were used to transform near-isogenic lines (NILs) of QPM. By using QPM germplasms as the donors of o2 gene and common germplasms as the receptors, QPM NILs were constructed by hybridization transduction. SSR marker phi057 was utilized to select QPM NILs, which could detect pure recessive o2 gene. The results showed that the co- dominant marker phi057, which could distinguish different genotypes O2O2, O2o2 and o2o2, had polymorphism among donor inbred lines and receptor inbred lines; the lysine content of QPM NILs (which came from different inbred lines) was increased in different levels, and it could reach 0.36%-0.42%, different receptors increased lysine content ranged from 9.1% to 64.0%. After introducing o2 mutant gene of the donors, the lysine content of the receptors was increased.
