Molecular Cloning and Bioinformatics Analysis of SFB Genes Controlling Self-incompatibility in Xinjiang Wild Almond (Prunus tenella Batsch.)

Abstract: The study aims to clone full-length sequence of the new pollen SFB gene of self-incompatibility in Prunus tenella. Xinjiang wild almond was used as the material, and molecular homologous cloning and RT-PCR were conducted in this study. The online software was used to analyze the protein physicochemical properties of SFB. The full-length gene sequences of PtSFB5 and PtSFB6 were successfully cloned. The full-length sequence of the two SFB genes was 1124 bp and 1072 bp, respectively, and encoded 374 aa and 348 aa, respectively. Both of the two genes had F- Box structure, belonged to the F- Box gene family. The predicted relative molecular mass was 30787.26 and 44037.74, and the isoelectric point was 5.93 and 5.43, respectively. PtSFB5 and PtSFB6 protein belonged to the unstable water-soluble protein.
