Abstract:

Using paraffin section and anatomical observation techniques, the process of leaf bud and flower bud initiation and flower organ differentiation of Daphne giraldii was studied, and the characteristics of leaf bud growth and flower bud differentiation of D. giraldii were clarified, so as to provide theoretical basis for D. giraldii breeding and systematic classification research. The results showed that: (1) the leaf buds of D. giraldii were distributed at the top and both sides of the old branches, which were divided into terminal buds and leaf buds. The entire leaf bud was conical in shape with scales covering the outside, the top being convex, and unconspicuous stipules. The leaf primordium differentiation period of leaf buds was from mid-late March to mid-April. (2) The flower bud differentiation of D. giraldii could be divided into 7 stages (mid-April to mid-May) including undifferentiated stage, early stage of differentiation, inflorescence primordium differentiation, floret primordium differentiation, petal primordium differentiation, stamen primordium differentiation, and pistil primordium differentiation. (3) A new stem was sprouted from the center of the leaf bud of D. giraldii (mid-April), and the flower bud grew at the apex of the central stem tip of the new stem from the leaf bud. (4) The observation of internal structure showed that the time of new stem emergence and flower bud differentiation of D. giraldii was synchronous.
