Effect on the Tissue Structure of Partridge Shank Chicken’s Spleen with Different Dosage Extraction

Abstract:

To study the effect on the tissue structure of Partridge Shank Chicken’s spleen with different dosage extraction from Pilose Asiabell Root and Herba Epimedii Brevicornus. 420 Partridge Shank Chickens of 14-day-age were randomly divided into seven groups. Control groups and experiment group were drunk water and different doses of extract from Pilose Asiabell Root and Herba Epimedii Brevicornus, Setting the Ⅰ group as contrast; setting the Ⅱ, Ⅲ, Ⅳ and Ⅴ,Ⅵ,Ⅶ group as small(0.5 g/L), middle(1.0 g/L), large(2.0 g/L) doses of extraction from Pilose Asiabell Root and Herba Epimedii Brevicornus. In 21th, 28th, 35th, 42th, 49th and 56th day morning, select 12 chicken for dissection before they were fed, taking anatomical specimens quickly and keeping it under Bouin liquid in normal atmospheric temperature 24 hours to be fixed, then, Dehydrated by ethanol, Make it transparent by xylene, paraffin embedding, Cutting it into 5um tire-road friction coefficient with the LEICA-RM-2128 type slicing machine, HE dying, sealing in piece with neutral gum, OLYMPNS-CH30 microstructure for observation, making the microstructure photography with microscopic photography system. The result show that experimental group of Pilose Asiabell Root and Herba Epimedii Brevicornus chicken spleen red pulp and white pulp had a clear boundaries, spleen bodies is obvious, the number of sheath which around arterial has increased, periarterial lymphatic sheath thickening. The 2 g/L group is obvious. The results account for that Pilose Asiabell Root and Herba Epimedii Brevicornus can promote the development of the spleen.
