Abstract: Isolated microspore culture is also called pollen culture, which is a branch of the development of plant tissue culture dependent on totipotency of plant cell. Making use of this technology, we could obtain homozygote, shorten the period of conventional breeding, and provide vector or objective genes for other subjects such as genetic engineering and cellular engineering. This paper summarized the influential factors in isolated microspores of Brassica vegetables, such as genotype of thematerials, plants physical condition, developing phase of microspore, physiological status of donorplants, separation methods and pretreatment way, adding methods of hormones and activated corbon etc for lasted years. And the existing problems of the microspore culture in the research and expectations in the future development has been came up, combining with the practical situation.
