Study on the Maize Transformation with Haloxylon ammodendron BADH Gene by Agrobacterimu tumefaciens

Abstract: This study explored the effect of factors on Maize transformation efficiency and had important significance to establish and perfect the maize genetic transformation system. Taking the‘NingYu 0905’as the study material, the transcription factor BADH gene was transformed into Maize genome of‘NingYu 0905’by Agrobacterium tumefaciens. The results revealed that adding acetosyringone could improve the transformation efficiency in corn in darkness. The highest transformation efficiency in corn was obtained when OD600 Agrobacterium tumefaciens was 0.3 for 5 min. The best transformation efficiency in corn was Agrobacterium tumefaciens and callus in 2 d CO-cultivation. The BADH gene was proved to be transferred into maize genome by PCR test of transformed plants. The highest transformation efficiency in corn was detected in 2 d CO-cultivation after 0.3 of OD600 Agrobacterium tumefaciens for 5 min.
