Stem Segment Tissue of Sabina chinesis‘Lanta’: Paraffin Section Technology

Abstract: The paper aims to obtain the paraffin section preparation method for the cross section of the Sabina chinesis‘Lanta’stem, and lay foundation for further study on the mechanism of rooting anatomy of the cuttage. We selected 16 cm long stem of Sabina chinesis‘Lanta’, cut the base 5 mm as the material, and modified the traditional paraffin section, such as softening, sticking and dyeing. The results showed that: the softening schemes of 20% hydrochloric acid immersed for 24 h and boiled for 4 d were better than that of concentrated hydrochloric acid immersed for 24 h, boiled for 10 d and glycerol- ethanol soaked for 10 d; reasonable dehydration and transparent time was 2 h per level; the optimal paraffin section thickness was 10 μm; the traditional adhesive method was modified with the mixture of gelatin tablets and 40% ethanol; paraffin sections obtained by staining with safranin for 60 min and fast green staining for 10 s were clear and time saving. This study provides a set of specific operation steps for the paraffin section technique of Sabina chinesis‘Lanta’ stem, and also provides a reference for the study on similar tree species.
